Chinese Simmental and Angus cattle have good meat performance and are the main beef cattle breeds in Tongliao region of Inner Mongolia. The fat deposition of backfat is the main indicator to measure the nutritional value and production function of beef.In this paper,an independent sample T test of the meat measurement data was obtained from the slaughtering test. The results showed that there were significant differences in warner-bratzler shear, drip loss, cooked meat rate and back fat thickness between Chinese Simmental cattle and Angus cattle(p<0.01).Cluster analysis was carried out to select three Simmental cattle and three Angus cattle respectively. The transcriptome data were obtained from six backfat tissues through BGISEQ-500 platform. The results of sequencing showed that the average mapping ratio of samples to genomes was 92.32%,the average mapping ratio of samples to genomes was 80.69%.A total of 21,970 genes were detected,19,747 new transcripts were detected,of which 16,576 were predicted coding transcripts,15,643 were new alternative splicing subtypes of known protein-coding genes, and 933 were new protein-coding genes transcripts, the remaining 3,171 are lnc-RNA.GO functional classification notes 3,149 differentially expressed genes(DEGs)in biological processes,2204 DEGs in cell component and 890 DEGs in the molecular function database.Separate classification and enrichment analysis of the three major functional classes.By analyzing the KEGG metabolic pathway, the Unigene of bovine backfat tissue was involved in 6 major categories and 44 subcategories of metabolic pathways, and a total of 2158 genes were annotated. The differential genes were screened out and combined with the relevant data of the slaughter test. The accuracy of the transcriptome data was verified by qPCR,and the genes DKKL1,ACACB and PTGS2 were initially predicted to be genes related to fat deposition.
