The pollen of melon varieties CM10,L94,m29 and watermelon varieties C11,w44 and W54 were used as the main pollen materials in the vegetable institute of Shandong Academy of Agricultural Sciences.The activity of pollen was determined by iodine potassium iodide staining,and the germination rate of pollen was determined by pollen culture in vitro.1.In vitro culture medium of muskmelon pollen,15% sucrose and 0.02% boric acid were the most suitable for 2 hours,while in vitro culture medium of watermelon pollen,15% sucrose and 0.03% boric acid were the most suitable for 2 hours.2.The highest pollen activity of m29 and w44 was at 9 a.m.at this time,the pollen activity of muskmelon was 94.57%,the germination rate was 94.25%,the pollen activity of watermelon was 92.41%,and the germination rate was 91.92%.After nine o’clock,the longer the time,the lower the pollen activity.3.The lower the temperature,the longer the pollen storage time.The pollen of Muskmelon m29 can be stored for four days at 25 ℃,five days at 4 ℃,six days at 0 ℃,one month at-20 ℃ and two months at-80 ℃.W44 pollen can be stored for two days at 25 ℃,four days at 4 ℃ and five days at 0 ℃.Therefore,pollen inactivation is faster at room temperature and should be used as soon as possible.4.For the growth and development of fruit after pollination,the longer the storage time is,the smaller the individual is after the fruit ripens.The average fruit weight was 0.8kg after one day of pollen pollination at25 ℃,and 0.45 kg after four days of pollen pollination.The average fruit weight was 1.2kg after one day storage at 4 ℃,and 0.7kg after four days storage.The average fruit weight was 1.3KG after one day of storage pollen pollination at 0 ℃,and 0.5kg after four days of storage pollen pollination.5.The pollen of m29,a melon variety with different developmental stages,was collected in advance and stored in low temperature before pollination.The fruit setting rate was as high as 95%.The unsealed male flowers picked 36 hours,24 hours and 12 hours in advance can be preserved at 0 ℃ and 4 ℃,and can be pollinated artificially after reaching the predetermined flowering time.6.For the pollen of Muskmelon Variety m29 in different pollination period,the activity of pollen in the early stage of pollination is the highest,the activity is 94.50%,the germination rate is 95.25%,the second is the activity of pollen in the middle stage of pollination,the activity is 89.20%,the germination rate is 95.09%,the activity of pollen in the late stage of pollination is the lowest,the activity is 84.87%,the germination rate is 86.34%.The pollen activity of w44 was the highest in the middle stage of pollination,with 90.30% activityand 92.57% germination rate.Secondly,the activity of pollen was 85.02% and the germination rate was87.11%.In the later stage of pollination,the activity of pollen was the lowest,the activity of pollen was87.45%,and the germination rate of pollen was 85.09%.7.There was no significant difference in the activity between the male pollen of melon varieties CM10,L94,m29 and the complete pollen,and there was no significant difference in the activity between the male pollen of watermelon varieties C11,w44,W54.This study can provide reliable scientific basis and technical data for the research of hybrid breeding and flowering regulation of melon,accelerate the breeding process and improve the conservation of planting resources. |
