Cefquinome is the first fourth-generation cephalosporin antibacterial agent used only for veterinary,as a broad-spectrum antibiotics of p-lactam,Cefquinome has the characteristics of wide antibacterial spectrum,rapid absorption,strong antibacterial activity and low residue.Compared with the previous three generations of Cephalosporin antibacterial agents,Cefquinome has an extremely strong killing effect on various clinical drug-resistant strains.Considering the poor absorption in the digestive system,Cefquinome was gradually getting alkylation to make its hydroiodate,after being deionizing,and reacted with sulfuric acid to make its sulfate,called Cefquinome sulfate which improved its stability as well.Cefquinome sulfate have good effects on bovine mastitis and endometritis,swine respiratory syndrome and meningitis and other bacterial disease which caused by E.coli,salmonella,drug-resistant staphylococcus aureus,etc.With the widely used in veterinary clinic,the detection method of Cefquinome sulfate by high performance liquid chromatography(HPLC)was established in in this research,and the elimination regularity and residues of Cefquinme sulfate were determined as well,which provides scientific basis for the safe and rational use of antibiotics.To understand the elimination of residue and the withdrawal time of Cefquinome sulfate in eggs,the detection method of Cefquinome sulfate by HPLC was firstly established.Based on relevant references,the mobile phase was determined as 0.05%acid solution-acetonitrile(120:10,V/V),after being extracted,the extract solution was cleaned up on C18 solid-phase extraction cartridges,the analyte was detected by HPLC under UV absorptive spectroscopy detector at 270 nm.According to the results,the average recoveries were ranged from 85.6%to 90.4%,and the coefficient of variances were ranged from 6.59%to 10.47%,which all within the prescribed scope.The data showed that the calibration curve was linearity at a range of 0.02-2.00 ?g·mL~-1;the lowest limit of detection(LOD)and the lowest limit of quantitation(LOQ)was 0.01 ?g·mL~-1 and 0.02?g·mL~-1 respectively.The stability of the sample was good and there was no interference of other homologous antbiotics.The chromatogram peak shape was good with high separation degree,indicating that the established method have good specificity and high accuracy.Based on the pharmacokinetic results and the clinical recommendation dose,the tested hens were subcutaneously injected at the dosage of lmg·kg~-1,one time dayly on the neck and continued for 12 days.The residual of Cefquinome sulfate in eggs was quite low.The maximum residues of Cefquinome sulfate was detected at the first withdrawal day in eggs,and the residues were reducing with the withdrawal time.At the third and the 5th of withdrawal day,the residues were lower than the lowest LOD and LOQ respectively.According to the MRL set by EMEA,the concentration-time dates were analyzed by WT14 software to get the withdrawal time with 95%upper tolerance limit,and the withdrawal time was advised for 2 d.The result demonstrate that the absorption and the elimination of Cefquinome sulfate is quick in hens,and the residues level is extremely low.It is proposed that the withdrawal time is short.
