Abstract/Summary:
The flowers of Aspidistra genus are small and flowering period is short,easily concealed,difficult in collection and has the large variation,classification and identification is difficult.So far,the classification system of Aspidistra is not satisfactory,although used cytotaxonomy,micro morphology,palynology and DNA barcoding identification of the plants of Aspidistra,but the effect was not ideal,which would seriously affect the in-depth study exploration and utilization of the medicinal plant of the genus.In this study,total DNA was extracted and purified from 25 species of the Aspidistra genus in China,we used Illumina high-throughput sequencing technologies to sequence and joint the contigs,and has got 25 complete chloroplast genome sequences.The complete chloroplast genome was annotated successfully and obtain chloroplast genome physical maps.We analyzed structure,composition,species of the chloroplast genome respectively,which laid a foundation for the identification and research of the Aspidistra genus medicinal plants.Detecting SNP(single nucleotide polymorphism)and Indel site of genome sequences,eomparing the karyotypes of Aspidistra genus plants genome polymorphism,results showed that there were variability in the whole chloroplast genome among the species of this genus.It was calculated and analyzed the interspecific genetic distance of the genus,and the results showed that phylogenetic relationship was closer of the species with smaller interspecific genetic distance,which were consistent with the phylogenetic analysis results.Using the maximum likelihood method,maximum parsimony,bayes method to construct the system of Aspidistra genus medicinal plant phylogenetic tree,the results showed that chloroplast genome sequences can successfully identify different species in the Aspidistra genus 25 plants.But there were differences in the traditional taxonomy of the genus.This study will construct efficient,rapid identification system of the Aspidistra genus,which provided the reference for the new method and new ideas for the medicinal plant DNA barcoding study.Which promote the solution to the problem of taxonomy of the genus and provide theoretical basis for the establishment of the infrageneric classification system.
