| 60 female Huainan partridge ducks(Hpds)with 40-day-old were raised in temperature for 20±1℃ 7 days.At the 8 day,all of the Hpds were divided into 6 groups and 10 Hpds in each group,and the control group was placed in 20±1℃,group 2、4、6、8、10h were subjected 40±1℃ acute heat stress,respectively.All of the Hpds were slaughted after heat stress treatment,the whole blood serum were taken to measure the biochemical parameters and blood physiological;the samples of heart and liver were fixed in 4%paraformaldehyde solution for paraffin tissue slides,the samples of heart and liver were placed in liquid nitrogen to detect the expression of HSP70 mRNA.The experiment results showed:The results indicated that the contents of serum TP,ALB and GLO in Hpds serum was significantly decreased by Heat stress(P<0.05).A significant increase in the activities of serum GPT which had a significant linear correlation with heat stress(P<0.05),The serum AST and CK activity increased with the time passing of heat stress,and peaked at 10h,which was significantly higher than that of other groups(P<0.05).The major pathological changes of Hpds under acute heat stress were the vessels between myocardial filled with erythrocyte,fiber clearance widens and splitting of myocardial fibers,necrosis and irregulation of tissue structure in liver.At 6h and 8h,the changes were most serious.The HSP70 mRNA expression of liver and heart was increased at first and decreased subsequently,and peaked at 6h and 4h,respectively.The conclusions show that Hpds changes the HSP70 expression and regulate the metabolism to response the influence of heat stress.The results of the study can provide evidence to healthful aquaculture the Hpds. |
