Phytophthora sojae is a highly destructive plant pathogenic oomycete.The soybean blight caused by it is a very devastating soil-borne disease in soybean cultivation.In order to explore the molecular mechanism of P.sojae,this study selected a gene Ps HSFs L that may be involved in regulating the germination of P.sojae based on the results of the transcriptome sequencing,and 2 plasmid vectors for transient silencing and overexpression of the gene were constructed and transferred into P.sojae by using PEG-mediated protoplast transformation scheme;Candidate transformants were selected through a medium containing G418 resistance,and then use quantitative verification,2 silencing and overexpressing transformants of the gene were selected,respectively.After that,we observed the growth phenotype and micromorphology of these 4 transformants,and studied their pathogenicity to determine the function of the gene in P.sojae and draw the following conclusions:1.Silence and overexpression of the gene Ps HSFs L have no significant effect on the morphology and size of P.sojae sporangia,but the silencing of this gene will cause bifurcation and deformity of the morphology of P.sojae sporulation.2.Silencing of the gene Ps HSFs L will reduce the activity of the extracellular protein laccase of P.sojae,while overexpression of the gene Ps HSFs L will significantly increase the protein and laccase content of P.sojae.3.The results of stress conditions on the growth rate of transformants showed that the silencing and overexpression of the gene Ps HSFs L have no significant effect on the mycelial growth of P.sojae.4.By comparing the pathogenicity of wild type and these 4 transformants on soybean plants,soybean leaves,and soybean sprouts,it can be seen that the silencing of the gene Ps HSFs L can significantly reduce the pathogenicity of P.sojae.In summary,we have confirmed that this gene is related to the proper formation of dormant spores of P.sojae,and is involved in regulating the pathogenicity of P.sojae to soybean.This confirms the importance of cell wall proteins of P.sojae in pathogenicity.It provides a theoretical basis for studying the action mechanism of these proteins during the infection of soybean by P.sojae,and also provides new research ideas and methods for studying the pathogenic mechanism of P.sojae.
