Xylaria is a dominant fungus on termite waste nurseries.Termites are currently one of the main pests that harm lawns,trees,architectural gardens,and other vegetation organisms.They not only cause major damage to garden vegetation,but also affect the construction of gardens and other buildings in cities.Therefore,the termite control in gardens is an important direction of garden protection.In this project,we sequenced the whole-genome and analyzed the secondary metabolites of Xylaria Escharoidea FB strain.The secondary metabolites of fermentation broth and mycelium were analyzed by LC-MS.The insecticidal and antibacterial activities were also bioassayed.The main results are as follows:1.The fruiting bodies collected in the field were isolated.The NCBI alignment of the sequencing results showed that the ITS sequence of the FB strain was 99% similar to the sequence of Xylaria escharoidea EU179864.1.The morphological characteristics of the fruiting body of the strain FB and the observation results of the electron microstructure are consistent with the characteristics of Xylaria escharoidea.The strain FB is determined to be Xylaria escharoidea.2.This whole genome of strain FB was sequenced.Sequencing data showed that the total number of bases in the FB genome was 8504443518 bp,the base recognition accuracy rate was as high as 99%,and the GC content was 51.39%.The low quality sequences were filtered out and deleted from the original data.Then the clean data was used for assemble,genetic prediction,and functional annotation.The results showed that there was a 10442 protein coding sequence in the whole genome of strain FB,the total length of the gene was 14129422 bp,the percentage of the gene in the genome was 40.7%,and the average length of the gene was 0.300 gene/kb.Functional annotation results show that the gene products of this strain are mainly biosynthesized.The prediction of secondary metabolites found 32 homologous gene clusters.3.The fermentation broth of the FB strain was tested for insecticidal and antibacterial activity.The results showed that the fermentation broth had a better antibacterial effect on Staphylococcus aureus.However,no insecticidal activity against Plutella xylostella was detected.The secondary metabolites of fermentation broth and mycelium were identified by LC-MS method.By comparing the literature,eight secondary metabolites were obtained,which were ?-linolenic acid propionate,cerebroside C,ergosterol,Fumagillin,3-carboxylic acid,chaetocin,allantoin,butyric acid,which mainly have antibacterial activity,anti-drug resistance activity,nematicidal activity,etc.
