In recent years,import of soybeans has been increasing continuously,and in which transgenic(genetically modified)soybeans accounted for a large proportion.Transgenic soybeans and their products could be seen everywhere in markets.Therefore there might be risks of contamination on non-transgenic soybeans in China.In this study 706 soybean varieties or breading lines were collected and detected qualitatively with PCR for the transgenic components.PCR detections were carried out on the 706 soybean varieties or breading lines for regulatory elements of CaMV35 S promoter,FMV35 S promoter and NOS terminator that mingt have be incorperated into or contaminated soybeans,as well as for the target gene EPSPS contained in the main herbicide tolerant transgenic soybean event GTS40-3-2 and MON89788,GM soybeans approved to be imported in China.At the same time,we designed EPSPS primer sequence to amplify 381 bp target band and established to detect MON89788 soybean,This method can be applied to the specific detection of MON89788 soybean.In order to establish a specific qualitative PCR detection method for MON89788 The 5'and 3' flanking sequence primers were designed to amplify 445 bp and 340 bp PCR products,which can be applied to the specific detection of MON89788 soybeans.Detection results showed the proportion of transgenic components containing Ca MV35 S promoter was 1%,FMV35 S promoter 0.85%,NOS terminator 0.71%,EPSPS target gene 0.85%,and herbicide tolerant transgenic event MON89788 0.85%,respectively.The evaluation was based on the above results,and provides data support for agricultural genetically modified regulatory authorities to carry out genetically modified soybeans monitoring.The uncertainties to the promotion and application of genetically modified crops are their harmful effects,which has plagued the development of genetically modified crops.In this study,based on the non-target metabolomics of UPLC-QTOF/MS,the differences of metabolites in roots,stems and leaves of transgenic soybean line GE-J16 and non-transgenic counterpart soybean Jack were analyzed.Fifty five different metabolites(VIP>1,P?0.05)were detected from the transgenic treatment group GE-J16 and the control group Jack,In soybean roots,the contents of?-linolenic acid and cellobiose changed greatly in ESI positive ion mode,and sorbitol changed significantly in ESI negative ion mode.In soybean stems,the contents of 1-aminocyclopropane carboxylic acid and hydrocortisone changed greatly in ESI positive ion mode In the negative ion mode,histidine changed obviously,but in the soybean leaves,there was no change in the content of compounds.The analysis of metabolites showed that the metabolites difference between transgenic soybean GE-J16 and non-transgenic counterpart soybean Jack was within the range of natural difference,and there was no harmful effect.
