Cucumber(Cucumis sativus L.)is an annual climbing herb of the cucurbit melon genus.It is one of the main vegetable crops widely cultivated in my country.As one of the most direct appearance qualities,the color of cucumber fruit peel is not only the main evaluation index for judging the ripeness of the fruit,but also an important factor to attract consumers.During the breeding and cultivation of cucumbers,people will choose valuable appearance traits such as fruit peel color,and increase the diversity of horticultural vegetable crop products through the use of target genes or beneficial mutations to meet the needs of consumers at different levels.demand.At present,the genetic research of cucumber fruit color mainly focuses on the white and green aspects of the young peel,and the study of the mechanism of the genetic development of the mature peel color can lay an important theoretical basis and practical guidance for the breeding of cucumber peel color.To this end,this study screened cucumber materials with different peel colors,measured the color difference of mature peels,analyzed the relationship between the peel color and the measured value of the color difference meter,and determined that the yellow-brown cucumber variety ’PW’ was the main experimental material.At the same time,using metabolome and transcriptome sequencing technology,GO functional annotation and KEGG enrichment analysis,identify the differential genes related to the color change of the peel during the development of the peel,and select the important structural genes and transcriptional regulators that affect the color change of the peel.And verify the expression of related qRT-PCR.The results are as follows:1.In this study,19 cucumber materials with different peel colors were screened to determine the color difference of the mature peel,and the relationship between the peel color and the measured value of the color difference meter was analyzed.The color difference analysis of 19 cucumber ripe peels showed that the change in peel color was significantly correlated with the color difference index L,a,and b.According to the color difference value,the ripe cucumber peel was divided into four colors:yellow brown,yellow,white,and green system.2.Using yellow-brown cucumber variety ’PW’ as the main test material,through metabolome and transcriptome sequencing technology,GO function annotation and KEGG enrichment analysis,27 differences related to the color change of the peel during the development of the peel were obtained Genes;further analyzed by WGCNA,screened 6 structural genes related to flavonoid anabolic pathway and 4 transcription factors,which may play an important role in the regulation of peel color changes;3.According to the analysis of differential metabolites of the metabolome,the genes involved in naringenin-chalcone biosynthesis were screened and related expression verification was performed.The candidate gene qRT-PCR analysis showed that CsGy3G024650(chalcone synthase),CsGy6G011750(Phenylalanine ammonia lyase),CsGy3G031120(4-coumaric acid-CoA ligase)and CsGy5G016860(chalcone isomerase)expression levels are closely related to changes in peel color,these genes are directly involved in naringenin-The biosynthesis of chalcone affects the color change of yellow brown cucumber peel.
