Objective Relating to the genetic information and medicinal ingredients synthetic gene function and metabolic pathways of Zhe-Maidong(Ophiopogon Japonicus(L.f.))from Tonglu and Cixi,and the defferences between them.Methods In Tonglu and collect biennial Zhejiang cixi in Zhe-Maidong gardens,dwarf lilyturf root extract Total RNA,construct a cDNA library and HiSeqTM-2500 double terminal sequencing,data filter assembly,will receive the Unigene and NT,NR,KEGG,Swiss-Prot,GO,COG,and compare the KOG database,analysis the differences in gene expression,SNPS and SSR analysis.Results Get 13.97 G original data,through data filtering and transcriptome spliced assembly,get Unigene 111736,total length of 83044647 bp,the average length of 743 bp and 1195 bp N50 length,will receive the Unigene and NT,NR,KEGG,Swiss Prot,GO,COG and compare the KOG database,respectively 51120,62634,38882,62634,34979,26816,article 44205 Unigene success ratio to the database.Will get to GO Unigene functional classification,divide 34980 Unigene success to three categories:56 functional group categories.Article 128 the metabolic pathways,including Zhe-Maidong main medicinal components flavonoids synthesis related metabolic pathways of article 5,saponin synthesis related metabolic pathways article 5 and polysaccharide synthesis related metabolic pathways in article 5 Found Zhe-Maidong SNP variation sites 252927,including Transiton has 163813,accounts for 64.77%of total variance;trransition has 89114,accounts for 35.23%of total variance.Found simple repetitive sequence 20863,distribution is one of the most double bases repeat sequence:7274,are three bases in after repetitive sequence:6316 and single-base repetitive sequence:5519.By gene expression difference analysis,130 differentially expressed genes were obtained,involving 23 genes functional classification and 48 metabolic pathways.Conncclusion there are significant differences between the Zhe-Maidong from Tonglu and Cixi.on transcriptomics.Transcriptome can be applied to study regional differences of Zhe-Maidong.
