In this paper,the non-steroidal anti-inflammatory drugs NS398 was used to treat the wound infection model of Streptococcus pyogenes.The clinical therapeutic effect of NS398 was evaluated by monitoring inflammatory cells,inflammatory factors,granulation tissue,fibroblasts and angiogenesis in dogs blood during drug treatment.Twenty healthy dogs were selected to establish a model of skin wound with S.pyogenes infection.NS398,cephalexin,penicillin and amoxicillin were selected in vitro with drug sensitivity test in vitro to determine the bacteriostatic effect of NS398 on Streptococcus pyogenes in vitro and the screening of control drugs.The NS398 were determined with antibacterial and screening in vitro.The dogs infected with Streptococcus pyogenes were treated with NS398 and cefalexin.The inflammatory cells,inflammatory factors IL-6 and TNF-? in the canine blood were monitored during the healing process by visual examination of wound healing.Monitoring of alpha and MCP-1,as well as granulation tissue,fibroblasts,and angiogenesis,were evaluated for the eveluation of clinical efficacy of NS398.The showed that the skin crescent-shaped incision was 1 cm×1.5 cm in length and the wound incision reached the depth of the dermis.Streptococcus had the best effect and the modeling success rate is 80%.Drug sensitivity test: After drug sensitivity test by NS398,cephalexin,penicillin and amoxicillin in vitro,NS398 had the best inhibitory effect on S.pyogenes it is butter than cephalexin.Further clinical trials were conducted with cephalexin as a control drug.The results of NS398 clinical treatment showed that the number of white blood cells,red blood cells and hemoglobin in blood of dogs in NS398 treatment group,Cefalexin treatment group and solvent group reached the peak at 24 hours before suppuration,0 h,24 h,48 h,72 h and 96 h after suppurative infection.The number of white blood cells decreased 48 hours after treatment.There was a significant difference between NS398 group and Cefalexin group and solvent group.The three indexes of NS398 group reached the standard range at 96 h.For the precise measurement of wounds at 0 h,24 h,48 h,72 h,and 96 h,the results of the healing rate showed that the cephalexin group increased significantly during the period of 0-48 h,and increased slightly during the 48 h period.The skin healing rate continued to increase steadily.After 48 h,it was significantly higher than that of the cephalexin group.After 96 h of treatment,the healing rate of the NS398 group and the cephalexin group approached 100%.After 3 days,7 days,and 12 days of infection,histopathological analysis showed that the inflammatory cell infiltration was severe at 3 days after infection,and fibroblast proliferation and angiogenesis were observed after 7 days of treatment.After 12 days of treatment,the drug was administered for 12 days.The granulation tissue regeneration was completed,and the fibroblasts were arranged neatly;the wound closure of the NS398 group was higher than that of the cephalexin group.The monitoring results of inflammatory factors il-6,tnf-alpha and McP-1 in the blood of the affected dogs showed that the values of inflammatory factors in the treatment group reached the highest after 48 h,and the values of inflammatory factors in the treatment group decreased to the lowest after 96 h,and the changes were most significant in the treatment group NS398.It can be concluded that NS398 has a higher healing effect than cephalosporin and has a good effect on skin healing.The histopathological analysis of the drug after infection for 3d,7d,12 d showed that the inflammatory cell infiltration was severe after 3 days of infection,and fibroblast proliferation and angiogenesis were observed after 7 days of treatment.The granulation tissue regeneration was completed after 12 days of treatment.The fibroblasts were arranged neatly;the wound closure of the NS398 group was higher than that of the cephalosporin group.The results of monitoring the inflammatory factors IL-6,TNF-? and MCP-1 in the blood of the dogs showed that the inflammatory factors were lower in the three experimental groups before the bacterial infection;after 48 hours of infection,three.The values of the test group reached the highest value;after 96 hours of infection,the values of inflammatory factors in the three test groups all dropped to the lowest point in the whole course of treatment,and NS398 decreased most significantly during the whole course of treatment.The results showed NS398 had a good inhibitory effect on the skin wound inflammation of S.pyogenes infection;NS398 cloud promote the repair of fibroblasts and the regeneration of blood vessels,and the infection of S.pyogenes Skin wound healing had a good promotion effect;NS398 was superior to cephalexin in the treatment of skin wounds caused by S.pyogenes infection,so NS398 can be used as a substitute for common antibiotics in clinical applications.
