Berberine(BBR),also known as berberine,is an isoquinoline alkaloid.It has been reported that berberine has a variety of pharmacological effects,including antibacterial,anti-cancer,anti-oxidant and anti-inflammatory.Macrophages are innate immune cells that are widely distributed in the body and play different roles in various physiological and pathological processes.Oxidative stress is a phenomenon caused by the imbalance between the reactive oxygen species produced and accumulated in cells or tissues and the detoxification ability of biological systems to these reaction products.Inflammatory responses are a rapid and coordinated response to microbial infections and tissue damage that can result in migration of immune cells to infected or damaged sites.There is a correlation between the concentration of ?-hydroxybutyrate(?-HB)in the blood of cows and the incidence of abdomen dislocation.The study found that the perinatal period of dairy cows is a high incidence of abdomen dislocation.During this period,cows have a wide range of oxidative stress and inflammatory reactions,and the energy metabolism and immunity of the body have undergone great changes.Therefore,the study of the protection of oxidative stress and inflammatory response of bovine macrophages has a certain significance for the preliminary exploration of the abdomen dislocation of dairy cows.In this paper,berberine was used as the research object to investigate the effects of berberine on oxidative stress and inflammatory response of ?-hydroxybutyrate-induced bovine macrophage(BM).First,the effects of berberine and beta-HB(2.4 mmol/L)at different concentrations(5,7.5,10 micromol/L)on the activity of bovine macrophages were detected.The oxidative stress and inflammatory reaction models of bovine macrophages induced by beta-HB were established and drug intervention was carried out.Secondly,the levels of ROS,MDA,SOD and CAT in bovine macrophages were detected by the kit,and the expression of proteins related to Nrf2 signaling pathway was detected by western blot to explore the effect of berberine on ?-hydroxybutyric acid induced oxidative stress in bovine macrophages.The results showed that berberine can inhibit the production of ROS and MDA in bovine macrophages.Furthermore,the content of SOD and CAT in bovine macrophages can be significantly increased.At the same time,the expression levels of Nrf2 and HO-1 were up-regulated.The above results confirmed that berberine has a protective effect on oxidative stress induced by ?-hydroxybutyrate in bovine macrophages.Finally,the expression levels of IL-1?,IL-6 and TNF-? mRNA were detected by fluorescence quantification,and the expression of NF-?B signaling pathway-related proteins was detected by Western blot technique.The results showed that berberine can inhibit the expression of inflammatory cytokines IL-1?,IL-6 and TNF-? mRNA,and berberine inhibits the activation of NF-?B signaling pathway by down-regulating the phosphorylation of I?B and P65.The above results confirmed that berberine has a protective effect on ?-hydroxybutyrateinduced inflammatory response of bovine macrophages.In summary,this paper preliminarily confirmed that berberine has protective effects on ?-hydroxybutyrate-induced oxidative stress and inflammatory response in bovine macrophages.The above results can provide data reference for the study of berberine on oxidative stress and inflammatory response induced by bovine macrophages.At the same time,it provides an experimental basis for studying the prevention and treatment of berberine in the abdomen dislocation of dairy cows.
