The intestine of animals is the main place for nutrient absorption,and the open intestine is prone to inflammatory reaction stimulated by various external environments.And inflammation often triggers a series of changes in inflammatory,further induces oxidative stress,aggravates intestinal and related tissue damage.Therefore,inhibiting or preventing the occurrence and development of inflammation will be one of the excellent strategies for disease.The intestine of animals has a close relationship with liver function,which plays an important role in the absorption and metabolism of nutrients in the gut-liver axis.In that case,when observing the development of intestinal inflammation,the intestinal and liver tissues are often studied as a whole.Our team found that astaxanthin showed excellent anti-inflammatory and antioxidant properties in hepatocyte,but whether it has similar characteristics in animals still needs to be studied.In this study,the intestine and liver of mice was used as the inflammatory model to explore the effect of astaxanthin on gut-liver axis in mice with acute inflammation by investigating the effects of astaxanthin on inflammatory factors and antioxidant capacity in acute inflammatory mice and its relationship with NF-?B pathway.Forty Kunming mice were randomly divided into control group,astaxanthin group,LPS group and protection group.Astaxanthin group and protection group mice were fed astaxanthin for 15 days before LPS treatment,LPS group and protection group mice were treated with 1 mg/kg LPS for 3 hours.The results are as follows:(1)First,this experiment established a lipopolysaccharide-induced acute inflammation model in mice and determined the optimal dose of astaxanthin in this trial.Compared with other LPS-treatment groups,the secretion and expression of TNF-?,IL-6 and IL-1? in serum and ileum tissues were significantly increased in the LPS-treatment group of 1 mg/kg for 3 h(P<0.05).Compared with LPS groups,the secretion and expression of inflammatory factors in serum and ileum were significantly lower(P<0.05)or decreased respectively in astaxanthin group at doses of 25 mg/kg,50 mg/kg and 100 mg/kg.The results showed that astaxanthin could alleviate acute inflammation in mice,and the follow-up experimental scheme was selected.(2)When studying the effects of astaxanthin on body's acute inflammation,we found that compared with the LPS group,the secretion and the relative expression of inflammatory factors mRNA and the expression of NF-?B p65 protein in serum or tissues of intestinal and liver in the protective group were significantly decreased(P<0.05),indicating that astaxanthin played a positive role in gut and liver of acutely inflammatory mice by NF-?B signaling pathway.(3)While studying the effects of astaxanthin on antioxidant levels in intestinal and liver tissues,it is found that Compared with the LPS group,astaxanthin significantly increased the antioxidant enzymes activity and gene expression of SOD,CAT,GSH-Px and GCLC in gut and liver,and decreased the content of MDA(P<0.05),indicating that astaxanthin enhanced the antioxidant capacity in mice.(4)When observating the morphology of the intestine and liver of mice,it was found that compared with the LPS group,the intestinal mucosa was less damaged,the villus morphology was intact,the liver cell morphology was intact and the liver damage degree was low in the protective group.It suggested that astaxanthin could protect the integrity of the intestinal barrier and maintain liver cell function.In summary,astaxanthin can protect the morphological structure of intestine and liver in mice,enhance the antioxidant capacity of the body,inhibite NF-?B signaling pathway,reduce the expression of inflammatory factors,alleviate the stress damage of gut and liver from various aspect.It was preliminarily confirmed that astaxanthin intervention had protective effects on mice with inflammatory response induced by lipopolysaccharide,and its mechanism remains to be further studied.
