Pork is the main meat food for Chinese residents,and improving pork quality has always been an important target in pig production.In essence,pork quality depends on the development of muscle and adipose tissue and the interrelationship between the two.Studies have shown that excessive fat deposition reduces lean meat quality and affects pork quality and flavor.Too low a fat content in the muscle also causes a decline in the quality of the meat.In addition,the quality of pork is closely related to the quantity and volume of muscle fibers.Therefore,on the one hand,the study of lipid deposition can provide a theoretical basis for improving pork quality and improving the carcass lean rate of pigs;on the other hand,it can also improve the economic benefits of the pig industry,with important animal husbandry significance and scientific research application value.Vaccarin is a flavonoid triglucoside extracted from Wang Bu liu xing,which is a kind of flavonoid glycosides and a pale yellow granular crystal.Flavonoids have lipid-lowering and anti-inflammatory activity,can inhibit lipid accumulation,inflammation and oxidative stress in muscle,fat,liver and other tissues,inhibit insulin resistance.Vaccarin also has the effect of regulating islet balance,thereby effectively alleviating lipid metabolism abnormalities and related chronic metabolic diseases.Studies have shown that the flavonoid glycoside can reduce the blood glucose content and triglyceride content,and has potential medicinal value.Therefore,in this study,C57BL/6 mice fed with high fat were gavaged with vaccarin,observe and detect physiological and biochemical changes in mice,and the potential mechanism of action was further explored to improve the corpus callosum for pig fat deposition.Provides a theoretical basis for quality and treatment of obesity-related diseases,and also provides new ideas for the treatment of obesity and related metabolic diseases.The main results of this study are as follows:1.Compared with the control group,the mice treated with the vaccarin(25mg/kg/day)in the group could significantly reduce the weight of the gonadal fat,the weight of the perirenal fat(P<0.05)and reduce the weight of the subcutaneous fat,thereby reducing the body weight of the mice.Changes of food intake in mice often result in changes in body weight.Based on this,the study found that the food intake of the mice in the vaccarin(25mg/kg/day)treatment group was not significantly different from that of the control mice,indicating that the body weight of the mice was regulated and not caused by food intake.2.Compared with the control group,the treatment of vaccarin(25mg/kg/day)significantly decreased plasma glucose(P<0.05),indicating that vaccarin has the function of regulating glucose homeostasis.In addition,the level of triglyceride(TG)in the blood of the mice treated with vaccarin(25mg/kg/day)was significantly decreased(P<0.05),and the liver fat content decreased,indicating that vaccarin has reduced blood lipids and the potential to relieve liver fat deposition.3.Compared with the control group,the weight of the soleus muscle(SOL)in the vaccarin(25mg/kg/day)treated group was significantly increased(P<0.05),and the muscle endurance was enhanced,indicating that the vaccarin can regulate muscle development and exercise performance.4.Further studies showed that compared with the control group,vaccarin(25mg/kg/day)treatment increased the transcription level of lipolysis-related genes ATGL and HSL in mouse gonad adipose tissue and subcutaneous adipose tissue,and improved ATGL and HSL in gonadal adipose tissue.The expression level of the protein and the transcription level of the lipid synthesis related genes Fas,Ppar? and Dgat2 were significantly decreased(P<0.05).In addition to white adipose tissue,vaccarin treatment also up-regulated the transcription level of the heat-producing gene UCP1 in mouse brown adipose tissue.5.Compared with the control group,the treatment of vaccarin(25mg/kg/day)promoted the expression of IRS1 of the insulin signaling pathway in the liver.At the same time,in the skeletal muscle,the treatment of vaccarin(25mg/kg/day)in the muscle not only promoted the protein expression levels of IRS1 and PI3 K in the insulin signaling pathway,but also promoted the protein expression levels of the myogenic regulatory factors MyoD and MyoG,thereby promoting the development of muscle.In summary,vaccarin can reduce the accumulation of lipids in the body by down-regulating lipid synthesis and up-regulating the expression levels of lipid-related genes,and further regulating insulin signaling pathway-related genes to enhance insulin sensitivity;In addition,vaccarin can regulate the growth factor and increase the soleus muscle weight,promote muscle development and improve muscle performance.
