| Melatonin is a natural hormone with a wide range of biological functions,which can enhance immune and antioxidant capacity.It can not only promote the apoptosis of tumor cells,but also inhibit the apoptosis of normal cells.Melatonin is also resistant to viruses such as rabbit hemorrhagic disease virus,equine encephalitis virus,herpes virus and parasites such as trypanosoma,toxoplasma gondii and schistosoma japonicum.Hair follicle is a very complex structure and function of sub-organs,which is an ideal model for stem cell research and organ regeneration research.This research explored the regulatory effect of melatonin on apoptosis of hair follicle in wan strain angora rabbits.Different doses of melatonin were embedded in rabbits for a trial period of 70 days.The results are as follows:First: By comparing different doses of melatonin-treated angora rabbit hair weight,it was found that melatonin could weaken the hair follicle apoptosis of Angora rabbits in summer and increase the yield of wool.Second: Through H&E staining and TUNEL staining,it was shown that 55 mg melatonin treatment could effectively inhibit the apoptosis of hair follicle cells and maintain the normal morphology of cells.Third: Differentially expressed genes were obtained by sequencing the 55 mg melatonin treatment group and the control group.These genes were used to perform KEGG functional enrichment analysis.The results showed that they were involved in necroptosis and ferroptosis.There were 6 differentially expressed genes associated with apoptosis,including 3 up-regulated genes and 3 down-regulated genes.Fourth: Four apoptosis-related differentially expressed genes were selected for Realtime-fluorescence quantitative PCR verification.As a result,the expression of MSTRG.3561 gene was significantly up-regulated and the expression of SLC25A5 was down-regulated,but there was no significant difference.Fifth: The structural analysis and phylogenetic analysis of the new gene MSTRG.3561 using bioinformatics showed that the total length of MSTRG.3561 gene was 1068 bp,which encoded a protein of 298 amino acids.The content of G + C was higher than that of A + T.The weight of the MSTRG.3561 protein molecular was predicted to 32.9862 ku and isoelectric point(PI)was 9.75,as the hydrophobic protein.It was located in mitochondria,which contained two strong transmembrane regions and multiple important functional motifs.Homology analysis andphylogenetic tree analysis indicated that MSTRG.3561 was highly conserved and had a close genetic relationship to Oryctolagus cuniculus,which was consistent with the traditional animal taxonomy. |
