| Botrytis cinerea is one of the most destructive pathogens in the world.This pathogen has a wide host range that includes cucumber,tomato and strawberry.Trichoderma is the most useful biocontrol fungi which can control Botrytis cinerea.The study on the biocontrol mechanism about Trichoderma against Botrytis cinerea is the key field of Trichoderma research.However,there is a lot of unknown field about the interaction mechanism among Trichoderma-plant-Botrytis cinerea.So,based on the Trichoderma which can antagonism Botrytis cinerea and promote cucumber growth,the expression situation of hormone synthesis related gene,the amount of hormone,the analysis of transcriptome and proteome were determined in the interaction system among Trichoderma-plant-Botrytis cinerea.1.Trichoderma strain H9 which can antagonism Botrytis cinerea and promote cucumber growth was isolated through the dual culture method and hydroponic method.The H9 strain was identified as Trichoderma longibrachiatum based on morphology and gene sequence analysis.2.The interaction system of Trichoderma-plant-Botrytis cinerea was established according to the method that Trichoderma was inoculated into the root and Botrytis cinerea was inoculated into the leaves of cucumber.In this system,the biocontrol efficacy to gray mold disease were 49.62%、55.85%、56.70% at 4dpi、8dpi and 12 dpi respectively.In this process,the hypha of Trichoderma can colonization in the root of cucumber.3.The expression situation of hormone synthesis related gene include LOX1、AOS、LOX2、EIN2、ACC、PAD4 and the amount of JA,SA in the interaction system were determined.The results showed that all the hormone synthesis related gene detected were up-regulated expression and the content of JA,ET,SA in cucumber leaf were increased at 96 h during the interaction system.4.The results of transcriptome in the interaction system showed that all the synthesis gene related to jasonic acid,ethylene and shikimic acid pathway gene related to SA synthesi were up-regulated.5.The results of proteome in the interaction system showed that most protein related jasonic pathway and all the protein related ethylene were up-regulated.Only the PAL related protein expression inn the SA pathway are up-regulated.6.Based on the data of hormone content,gene expression situation,transcriptome and proteome analysis,we can deduced that the signal pathway in plant responsing to Trichoderma induce included the jasmonic acid / ethylene and salicylic acid pathway.The former was the main pathway in this process. |
