The mononuclear heredity can be stable for the good characters of the parents,and can provide the basis for the genetic breeding of edible fungi.Two varieties of Coprinus comatus “92” "96" were used as the experimental materials in this experiment.Two monokaryons of two varieties were obtained by protoplast mononuclear technique,and the mating type was analyzed by mating of mononuclear bacteria.The monokaryons obtained from the two cultivars were identified and analyzed by ISSR molecular marker technique and esterase isozyme method respectively.On the basis of this,the mononuclear was selected for further pairing hybridization.According to the hyphal growth characteristics and growth status of hybrid strains,a good hybrid strain was screened,and the ISSR molecule was used.Marker technique and esterase isozyme methods were used to analyze the genetic difference of the selected hybrid strains,which laid a foundation for the genetic breeding of Coprinus comatus.The main findings are as follows:1.By using protoplast mononuclear technique,21 mononuclear strains were obtained,of which 11 mononuclear strains were obtained from "92" variety and 10 mononuclear strains from "96" variety.The mating type factors of "92" strain of Coprinus comatus were A1B1,A2B2;the mating type factor of "96" strain was A3B3.2.Mononuclear identification and analysis showed that the parent of Coprinus comatus "92" was superior to the protoplast mononuclear strain in mycelium growth rate and growth,while the parent "96" was not completely superior to the protoplast mononuclear strain.The results of ISSR molecular marker showed that "92" parent was superior to protoplast mononuclear strain.The band loci of the protoplast mononuclear "CC1-6" of "92" were significantly different from those of their parents,while the other mononuclear strains were the same as their parents.Compared with their parents,the amplified bands of the protoplast mononuclear of "96" variety had parental bands,which proved that the genetic similarity rate was higher.The results of esterase isozyme showed that the protoplasm of "92" variety Compared with their parents,the somatic mononuclear "CC1-6" had the same mobility as their parents,the other mononuclear strains differ from their parents,and the "96" varieties showed the same mobility as their parents.3.The results of pairing hybridization showed that 40 hybrids were superior to their parents that selected among the 100 hybridized stranis obtained according to the growth speed and characters of hypha.The results of ISSR molecular marker analysis showed that there was no significant difference between hybrid strain and parent strain "92",and had the same locus with parent "96" strain.The results of esterase isozyme showed that the mobility of the hybrid strains was the same,and the mobility of the hybrid strains was 0.69 more than that of the parent strain "92".Compared with parent strain "96" of Coprinus comatus,hybrid strain The migration rate wasthe same as that of "96" strain.
