Dendrobium is one of the largest genera of orchidaceae,The germplasm resources of Dendrobium in Sichuan and Yunnan area accounted for 85% of the total.And it has great value on theory,ornamental and medicinal,so it is necessary to study the diversity of resources and its genetic background to provide reference value for the development and utilization of dendrobium resources and its depth study.Therefore,through the resource collection and cultivation management of dendrobium,the statistical morphological differences were investigated,and the coefficient of variation(CV)was calculated.Using spectrophotometer,HPLC and HPAEC technology is used to detect the content and the establishment of quality inspection method,to test the chemical composition of dendrobium resources.Then,the genetic background was studied by ISSR molecular marker.1.Through morphological statistics,the CV value of morphological index is large,plant length,number of stem,section length,leaf number,leaf width six traits CV were 53.33%,69.39%,48.71%,55.70%,51.62%,61.99%,show that the morphological variation of Dendrobium is rich,has the rich diversity in form.2.HPAEC and HPLC detection methods can be used to determine the chemical composition of dendrobium with high efficiency and accuracy,At the same time monosaccharide composition(Glu,Man,Rha,Ara,Xyl,Fru,Lac)and other chemical composition(Erianin,Syringate,Naringenin,Quercetin,Coumarin,Gallic acid,vanillin)can be measured effectively,and the determination method of chemical composition can provide reference for quality control of dendrobium.3.By the 21 kinds of dendrobium effectively analyzing chemical composition,significant difference in moisture content,polysaccharide content,and monosaccharide composition,other chemical composition also differences in composition and content.This can be seen form the chemical composition on the collection of dendrobium on the chemical composition diversity.Through similarity analysis and comparison of Chinese Pharmacopoeia varieties,This collection of Dendrobium parishii and Dendrobium longicormu Lindl were worthy of attention.Through the analysis of different algorithms of PCA,The results of chemical composition clustering of different algorithms are different.But it can also play the role of chemical composition identification.4.The ISSR molecular markers selected 9 primers UCB811,UCB819,UCB835,UCB844,UCB855,UCB884,UCB886,UCB889 and UCB890 were all amplified with a clear and stable band,which amplified 626 strips,571 polymorphisim and 91.3% polymorphisim.By calculating the genetic similarity,the genetic similarity coefficient can be divided into 6 classes when the genetic similarity coefficient is 0.3.
