Lilium duchartrei Franch.is one of perennial bulbous flower,which belongs to the Lilium of Liliaceae.It is also a lily in the wild that is unique to China.L.duchartrei has great ornamental value and medicinal value.The market's prospect is very broad.L.duchartrei grows on the wet and chilly highland meadows in the northwestern region of Sichuan and Yunnan,as well as in Guizhou and Tibet.Overexploitation has caused a dramatic decrease in the number of L.duchartrei.At presentthe studies L.duchartrei are focused on resource investigations and social karyotype analysis,there is a lack of complete research on the organizational culture system.In this paper,L.duchartrei scales were used as explants to study of many bulblets of lily could be induced in tube and some factors influenced the bulblet formation were researched in tube.Culture media for adventitious bud induction,adventitious shoot proliferation,and rooting culture,through the influences of the factors such as temperature,sucrose concentration,NAA concentration,NH4+-N:NO3-N-,and PP33333 concentration to differentiation and propagation of bud and bulb development,in order to establish is a vitro regeneration system of L.duchartrei.Conclusion as below:?1?For the explants of L.duchartrei bulb scales,the best disinfection scheme was disinfection with alcohol for 15 s,then 0.1%HgCl2 for 16 min,it's contamination rate was3.33%and the mortality rate was only 8.33%;the differentiation ability of outer layer scales was the inner side of the scale upwards>the scale insert upright;the best medium for adventitious buld induction is MS+0.5 mg/L 6-BA+1.0 mg/L 2,4-D+1.0 mg/L AC,the browning rate was only 13.33%,the medium supplemented with activated charcoal is MS+0.5 mg/L 6-BA+1.0 mg/L 2,4-D+0.1 mg/L AC,it can ensure a lower browning rate and a higher differentiation rate.?2?The best subculture medium for L.duchartrei plantlet is MS+0.5 mg/L 6-BA+0.2mg/L NAA,multiplication coefficient was 11.00.?3?The addition of sucrose facilitated the formation and expansion of bulbs.When the sucrose concentration was 60 g/L,rate of differentiation was 75.00%,fresh weight gain of the bulb was 4.82 times;when the NAA concentration reached 0.5 mg/L,rate of differentiation was 100.00%,the fresh weight of the bulb was increased 3.33 times;when the ratio of NH4+-N:NO3-N-is 60 mmol/L:40 mmol/L,that is total nitrogen is 100 mmol/L,rate of differentiation was 95.00%,bulb fresh weight gain 3.33 times;when the temperature was23±1°C,rate of differentiation was 96.67%,the fresh weight of the bulb increased 2.50 times;when PP33333 concentration is 5 mg/L,bulb fresh weight gain 3.87times.?4?The suitable growth medium for L.duchartrei is peat soil:vermiculite:perlite=1:1:1,and the final survival rate could reach 93.33%.
