Present study was to investigate the effect of Konjac oligosaccharides on the immune system of Schizothorax prenanti.Besides,we also studied the immune-regulatory potential of Konjac oligosaccharides by challenging Schizothorax prenanti with Aeromonas hydrophila.Schizothorax prenanti were fed with diets that contain 0.4%、0.8%、1.6% KOS respectively for 8 weeks.Then they were challenged with Aeromonas hydrophila and sampled at 6h、24h、72h、168h respectively.The serum ACH50、C3、CRP、Ig M、NO、i NOS、SOD、GSH-Px、MDA activity or content,immune-related genes(TLR5、TLR22、My D88、TRAF6、NF-k B、TNF-α、IL-1β、IL-8 、 IL-10 、 β-Integrin 、 TGF-β 、 HSP70 、 CD40 、 PKR)expressions in all immune-related organs were analyzed.Results are as follow:(1)0.8% KOS significantly increased weight gain and specific growth rate of S.prenanti.(2)1.6% KOS significantly enhanced serum ACH50 activity,C3、i NOS、NO content of S.prenanti.(3)0.4%、0.8% KOS significantly enhanced serum GSH-Px content,0.4%、1.6%KOS significantly decreased serum MDA content.(4)1.6% KOS activated TLR5,TLR22 signaling pathway and finally up-regulated pro-inflammatory cytokine.Beisdes,up-regulated anti-inflammatory cytokine,which indicated that S.prenanti possessed a relatively high immunological state.(5)Aeromonas hydrophila significantly promoted immune-related parameters and immune-related genes expression,1.6% KOS can regulated the infected S.prenanti immune response and maintain it at the normal level.Therefore,1.6% KOS can activate the complement system of S.prenanti through alternative pathway;1.6% KOS can make S.prenanti possess a relatively high immunological state by activating TLR5、TLR22 signaling pathway and up-regulating anti-inflammatory cytokine;1.6% KOS can regulated the infected S.prenanti immune response and maintain it at the normal level;KOS can protect S.prenanti from oxidative damage. |
