Nowadays China's economy has improved a lot,people are now having an eagerer desire for and greater demand of better animal foodstuff.For the sake of cost reduction,high-efficient production and effective treatment of livestock,people tend to overuse animal medicines in the mid of processes of production,such as enrofloxacin,which is widely used for better clinical effect and lower side effects.Overdose of animal medicines may contribute to livestock's toxic reaction and emergence of drug-resistance bacteria,even more pollution of environment and a big threat to human's health by faeces and food chains.Pork is the biggest consumption among sources of meat in China,however,there are limited studies on connection and mechanism of pigs and animal medicines.Liver plays a pivotal role in detoxification of organism,it would provide theoretical basis of its interaction with animal medicines to study the effects of that on liver.In the study,24 normal Guangxi Bama pigs(3~5-month-age,approximately 10 kg)were distributed to 4 groups(3 male and 3 female each)randomly and injected intramuscularly with normal saline as CK and different dose of enrofloxacin at 5(low dose group),25(middle dose group)and 125(high dose group)mg/kg·BW respectively for consecutive 5 days to determine the effects of enrofloxacin on pig liver proteome by applying 2-DE and mass-spectrometric detection,CYP450 enzyme system and antioxidant system by ultraviolet spectrophotometry.Results showed: 1.expression of GLOD4 was depressed in each group significantly(P<0.05)and HSP70 was depressed in middle and high dose groupsignificantly(P<0.05)in liver proteins extraction;Expression of liver carboxylesterase was induced in low and middle dose groupsignificantly(P<0.05),alpha-enolase was induced in high dose group significantly(P<0.05)in liver microsome proteins extraction.2.Content of CYP450 was induced gradually and reached a significant difference in high dose group(P<0.05);Content of CYb5 in each group was not affected obviously compared with control group;Activity of NCCR was induced gradually and reached an extremely significant difference in middle and high dose group compared with control group(P<0.01);Activities of AND and ERND were both depressed along with the increase of enrofloxacin and reached a significant difference in high dose group(P<0.05);Activity of AH in each group were not affected compared with control group.3.Activity of SOD in each group was not affected obviously while there existed a trend that activity went up and then down along with dose increasement;Activity of CAT was induced gradually and reached a significant difference in high dose group(P<0.05);Activity of GSH-Px was induced and reached a extremely significant difference in high dose group(P<0.01);Content of MDA increased very obviously in low,middle and high dose group compared with control group(P<0.01).Above all,overdose of enrofloxacin makes obvious impacts on proteic expression of pig liver,activities of CYP450 and antioxidant enzymes and content of lipid peroxide.Effect of quinolone antibiotics on other animals could also be evaluated based on the study.
