Anthocyanin synthesis is the most studied secondary metabolic pathway and related transcriptional gene regulation in plants.the anthocyanin pigment pathway was regulated by a suite of transcription factors that include MYB,bHLH and WD-repeat proteins,these three transcription factor form a ternary complex(referred MWB).Various transcription factor combinations can specify the class of flavonoid pigment produced,catalytic gene expression,to achieve the ultimate purpose of the regulation of anthocyanin synthesis.we use two eggplant varieties(‘hexian'and ‘ FGM ')to explore the regulation mechanism of anthocyanin biosynthesis in eggplant.1)The three anthocyanins synthesis related genes' cDNA,named SmTT8,SmGL3 and SmTTG1 were cloned from eggplant(Solanum melongena L.)cultivars ‘hexian ' using homology-based cloning method.RT-PCR analysis showed that the SmTT8 ?SmGL3 and SmTTG1 were expressed in all organs,including roots?stems?leaves?petals and peels.the expression level of SmTTG1?SmGL3 and SmTT8 were tissue-specific.2)Subcellular localization indicated that the SmGL3 and SmTT8 protein were localized in the plasma membrane and nuclears,the SmTTG1 protein was localized in the plasma membrane.Biomolecular Fluorescence Complementation(BiFC)showed that SmTT8 and SmGL3 were all interacted with SmTTG1 in the plasma membrane,and the yeast two-hybrid assays showed that SmMYB1 was all interacted with SmTT8 and SmGL3.3)The transcript level of the three genes were significantly upregulated exposure to the NaCl,ABA and 4? treatment.We examined nitrogen effects on expression of the three genes in the flavonoid pathway of eggplants,The SmGL3 transcript increased under nitrogen deficiency,However,the transcript level of SmTT8 and SmTTG1 were not significantly influenced by nitrogen deficiency.4)We bagged two eggplant varieties(‘Hexian'and‘FGM')to see how light influences transcription factors.The result showed that the SmTT8?SmGL3 transcript in the photosensitive type eggplant decreased under bagged treatment,but the SmCOP1 transcript was induced and the transcript level of SmTTG1 was not significantly influenced.However,the SmTT8?SmGL3 and SmTTG1 transcript in non-photosensitive type eggplant were not influenced by bagged,but the SmCOP1 transcription level were decreased decreased reduced after bagged.The different influence expression patterns between the regulatory genes in the non-photosensitive type eggplant is distinct with the photosensitive type eggplant,so we speculate it is relates with the regulation of light on anthocyanin synthesis.5)Use the sequence contrast to see the different gene transcription factors between the photosensitive type eggplant and the non-photosensitive type eggplant after bagged.we found that there were no differences between cDNA sequences and the promoter between the two type eggplant.
