In order to explore the expression of HIF-1? gene in different tissues of plateau yak of fetus.This dissertation selected plateau yak placenta and fetus as the research objects.The combination of histology,morphology and molecular biology were used to observe the placental morphology of yak during pregnancy,the expression of HIF-1?,PLGF in placental tissue and the expression of HIF-1? in different tissues of plateau yak of fetus.The expression of fetal tissues was explored to explore the adaptive characteristics and mechanisms of the plateau yak of fetus.The following is the main findings:(1)The increase of syncytial cell nodules,vascular syncytia membrane and blood vessels in the placenta of the four-month-old fetus was lower than that of Zeku yak of fetus.It indicates that the morphological structure of placenta has changed in order to adapt to the hypoxic environment at higher altitudes.(2)By comparing the expression of HIF-1? and PLGF in the placental tissues of embryos at 4 months of age,it was found that the expression levels of HIF-1? and PLGF in placenta tissue increased with altitude.It is suggested that in high altitude areas,in order to normal growth and development of the fetus during pregnancy,placental tissue can adapt to the hypoxic environment by increasing the expression of HIF-1? and PLGF.(3)The difference of somatic cell nodules,vascular syncytia membrane and vascular an increase in the placenta of Datong yak was not significant compared with Zeku yak(P>0.05),but the difference was extremely significant compared with the placenta tissue of Haiyan yak(P>0.05).It is indicated that the variety is also an important factor affecting the structure of placenta and the expression of HIF-1? and PLGF.(4)By analyzing the expression of HIF-1? mRNA and protein between plateau yak tissues at different altitudes,All of tissues have been discovered the gene,and the expression of HIF-1? was different(P<0.05),and it was higher in brain tissue.The amount of expression.(5)Long-term high altitude hypoxia adaptation can specifically express HIF-1? mRNA and protein between fetal tissues.In addition,long-term high altitude hypoxia adaptation results in higher expression of HIF-1? mRNA and protein at high altitude.In summary,the authors believe that the morphology of placenta,the expression of hypoxia-regulating factors in placental tissue and the expression of hypoxia-responsive genes during gestation are the adaptation to high altitude hypoxic environment.These studies have filled the gap of low-oxygen adaptation of plateau yak fetus,which is of great significance for enriching and perfecting the hypoxia adaptation of plateau yak,and is conducive to better protection of plateau biodiversity.
