Voltage-dependent anion channels(VDACs)are important bridges between mitochondria and cytosol,and play critical regulatory roles in transport,energy exchange,and cell apoptosis.In the previous study,7 interacting proteins were screened by yeast twohybrid using rice protein OsVDAC5 as bait,and named OsV5IP1-OsV5IP7.Subcelluar location and interaction vadication of OsVDAC5 and its interacting proteins were confirmed.In this study,transgenic plants of OsVDAC5 and its interacting proteins were used to research the functions of them through molecular identification and phenotypic check in agronomic traits and resistance to stresses.The progresses are as follows:1.RNAi plants of OsVDAC5 and T-DNA insertion mutants of OsV5IP5 and OsV5IP7 were identified in genotype,expression level and agronomic traits.Compared with the wild type ZH11,the OsVDAC5 RNAi plants showed lower fertility and setting rate as well as OsV5IP5 and OsV5IP7 homozygous mutants.The results suggested that OsVDAC5 and its interaction proteins may positively co-regulate pollen development.In addition,T0 generation of overexpression plants of OsVDAC5,OsV5IP5 and OsV5IP7 were identified by real time PCR and 4,5and 2 plants with up-regulated expression of the target genes were obtained,respectively.2.The expression profiles in stress and hormone treatments revealed that the expression levels of OsVDAC5,OsV5IP5 and OsV5IP7 were induced by multiple stresses and hormones.The expression level of OsVDAC5 increased obviously at the early of drought stress and ABA treatment,and then reduced to normal level,whereas it kept rising during salt,cold and SA treatment,but was not induced by JA treatment.Gene OsVIP5 showed down-regulated expressions in drought,salt and cold stresses,but up-regulated expressions in JA,SA and ABA treatments.The expression level of OsV5IP7 was down-regulated by drought stress,but up-regulated by salt,cold and SA treatment,and not influenced by JA treatment.Salt stress testing showed that loss of OsVDAC5,OsV5IP5 or OsV5IP7 enhanced the sensitivity of rice to salt stress,suggesting that OsVDAC5 and its interaction proteins may co-regulate the tolerance of rice to salt stress.In summary,rice OsVDAC5 and its interaction proteins may cooperatively play positive roles in pollen development and salt stress.
