As a kind of national second-class endangered plant,Psammosilene tunicoides is used as a root medicine and has the effect of analgesia and anti-inflammatory,etc.In recent years,with the continuous rise of market demand,P.tunicoides has suffered from predatory mining,so it is urgent to carry out seed seedling breeding of P.tunicoides in a timely manner.Because the existing seedling breeding technology is not mature enough,it is very important to protect the wild resources.In this paper,the sexual reproduction technology,in vitro propagation technology and industrialization base construction of P.tunicoides seedlings were studied.The main research findings were as follows:1.The sexual reproduction research shows that 4?is more suitable for long-term preservation of P.tunicoides;The seeds should be treated as follows before sowing:15%hydrochloric acid soaking for 30 min and then soaking in 200 mg/L 6-BA for 24 hours or soaking in 750 mg/L GA3 for 24 hours.After hormone treatment reached the highest germination rate and germination index.After 6-BA treatment the germination rate was the highest and the growth was the best and the germination rate was higher after soaking in GA3for 24 hours and the radicle growth was good.Different cultivation conditions have an important influence on the growth and the medicinal components of P.tunicoides seedings.The results showed that leaf area,number of leaves and internode distance tended first increase and then decrease with the increasing of shade.The fresh and dry weight of aboveground and underground part and the root-crown ratio were negatively correlated with the shading degree.The contents of total saponins were as follows:light shading>medium shading>full sun light>severe shading.With the increasing of fertilizer application,each index showed a change of first rising and then falling,and the total saponins content showed as follows:0.3 kg/m2>0.6 kg/m2>no fertilization>0.9kg/m2.There was no significant difference among the growth indexes under different substrate treatments.The contents of total saponins were as follows:field:substrate:river sand=2:2:1>field>field:substrate=1:1.Based on growth indexes and total saponin content,shading degree of 75%85%,fertilizer amount of 0.3 kg/m2,field:substrate:river sand=2:2:1 have positive effects on the growth and development of germchit.2.The results of rapid propagation technique showed that:the appropriate treatment of seed disinfection was:75%ethanol 30s,0.2%potassium permanganate 10s and 0.2%mercury15min,and the pollution rate was 0.The suitable seed induction method was soaked in 200mg/L 6-BA for 24 hours that germination rate was 57.33%.The suitable medium for budding induction was MS+2mg/L 6-BA+0.3 mg/L NAA+30 g/L sucrose+7 g/L anger power.The appropriate proliferation medium was MS+0.05 mg/L TDZ+0.8 mg/L 6-BA+0.2mg/L NAA+30 g/L sucrose+7 g/L anger power.The feasible rooting formula was:3/4MS+0.3 mg/L IAA+0.1 mg/L NAA+50 g/L banana juice+30 g/L sucrose+7 g/L anger power;The tissue culture bottle opening time was 3d,and the matrix formula was humus:perlite 2:1.3.The idea of P.tunicoides seedings industrialization base construction was preliminarily explored by using the method of school-land combination and complementary advantages,and the construction plan of P.tunicoides wild conservation and nurturebase in Weining county,Guizhou province was compiled in the main production area.
