| In order to optimize the rooting time and concentration of rooting agent,this experiment determined the cutting substrate and density,improved the utilization rate of seedlings,and shortened the cutting time.In the present study,early-maturing potato variety of ‘zaodabai’ tissue culture seedling and late-maturing potato variety of ‘Atlantic’ tissue culture seedling were used as research materials.Analysis in the laboratory,from cuttings rooting agent concentration time selection,cutting the choice of substrate,selection of cutting density,the choice of appropriate section number of potato seedling propagation as well as cutting the original seed technology is studied.The concentration and time of IAA with indoleacetic acid were selected,determination of survival rate,root vitality,root number,root length and root weight;screening the cutting matrix and cutting times,the plant height,stem diameter,leaf area,leaf number,POD,SOD,chlorophyll content,soluble protein content,minimum single tuber size,maximum single tuber size and total yield were measured;screening the cutting density,tuber number per plant,yield per plant and total yield.The main conclusions are as follows:1.Rooting agent indole acetic acid IAA,the effect is good,high survival rate.The most suitable treatment time was 25 s,the most suitable soaking concentration was 1.0mg/L.Both cultivars showed that the rooting time was shortened to 4d ~ 6d and the survival rate was above 99%.2.Vermiculite-peat mixed matrix can promote the agronomic characters,physiological indexes and yield indexes of potato cutting seedlings.Substrate S1 composed of vermiculite and peat increased the agronomic characters,physiological indexes and yield indexes of potato cutting seedlings to varying degrees.The plant height of two potato cuttings in S1 was 1.10~2.52 times higher than that of CK,the stem diameter was 1.01~1.64 times higher than CK,the leaf area was 0.65~0.72 times higher than that of CK,and the number of leaves was higher than that of CK.0~3 p;peroxidase increased 0.78~0.84 times compared with CK;superoxide dismutase increased 0.62~0.78 times compared to CK;chlorophyll content increased 0.53~0.78 times compared to CK,soluble protein content Compared with CK,it increased 0.41~0.97 times.At the same time,the total output increased 1.21~1.62 times that of CK.3.Potato tissue culture seedlings in the first four cut off with the mother plant difference is small,but after the fifth cut off offspring began to appear different degrees of degradation.Compared with the previous four cuttings,the maternal plants of two varieties of potato grew 0.1 cm ~ 5.9 cm in height,0.01 cm ~ 0.039 cm in stem diameter and leaf area increased by 0.49~0.75 times,leaf number increased by 1~5 p,peroxidase content increased by 0.18~0.46 times,superoxide dismutase increased by 0.47~0.61 times,and chlorophyll content increased by 0.18~ 0.31 times,the soluble protein content increased by 0.28-0.35 times while the output increased by 1.08~1.52 times;the height of the two varieties increased by 0.15~0.34 times compared with the fifth-cut segment,and the stem thickness increased by 0.0001cm~0.007 cm.The area increased by 0.48cm2~40.17cm2,the number of leaves increased by 1 ~ 6 p,the peroxidase content increased by 0.21~0.56 times,the content of superoxide dismutase increased by 0.53 ~0.69 times,and the content of chlorophyll increased by 0.20 ~0.43 times.The content of soluble protein increased by 0.21~0.45 times,and the yield increased by 1.37 ~ 1.82 times.4.Early white Daiche optimum cutting line spacing for the plant line spacing is 4cm × 5cm,density of 500 / m2,Atlantic most suitable cutting line spacing is 6.7cm × 5cm,that is,the density of 300 / m2.Compared with CK increased by 0.20 g ~ 0.51 g,The yield per plant increased by 1.03~1.47 times compared with CK,and the total yield increased by 1.26 ~ 1.85 times compared with CK. |
