Preparation Of Resveratrol Dry Suspension And Its Effect On Immune Function

Resveratrol(trans-3,4,5-trihydroxystilbene),a natural polyphenol compound mainly found in polygonum cuspidatum,peanuts,and grape skins,and exhibits a wide range of pharmacological activities.However,the poor water solubility and structural stability limited its application.This study prepared Resveratrol Dry Suspension(RDS)and evaluated its immunomodulatory and anti-inflammatory activities in mice.Subsequently,we studied the effects of RDS on immune function in piglets.1 Preparation of Resveratrol Dry Suspension and its quality evaluationSingle factor test and orthogonal factor test were used to screen the excipients suitable for the preparation of RDS.The HPLC method was used to determine the content of resveratrol in RDS.The indicators consisted of appearance,odor,identification,loss on drying,settling volume and redispersability were used to assess the quality of this preparation according to the Chinese Pharmacopoeia.2 The effect of Resveratrol Dry Duspension on the immune function of miceMice were injected intraperitoneally with CY to establish immunocompromised models The index of immune organs,lymphocyte proliferation,the ratio of CD3+CD4+/CD3+CD8+ and the expression of cytokines were used to reflect the immune function of mice.The anti-inflammatory effects were observed in mice by acetic acid increased capillary permeability,xylene-induced ear edema and cotton pellet granuloma.The results showed that the resveratrol dry suspension could increase spleen index,regulate T lymphocyte subsets,increase the level of IL-2,and improve the proliferation of splenic lymphocytes.Moreover,it can inhibit the acute inflammation including capillary permeability and ear swelling response,inhibit granuloma in chronic inflammatory reaction.Our study suggested that the dry suspension was a suitable preparation for resveratrol exerting immunoregulatory and anti-inflammatory activities.3 The effect of Resveratrol Dry Suspension on the immune function of pigletsThe experimental results of RDS on piglets demonstrated the good effect of promoting immune function.RDS has significant effects on the development,maturation,proliferation and transformation of T lymphocytes.RDS could regulate humoral immune responses by upregulating the release of IFN-? and downregulating the release of TNF-?.After piglets vaccinated against classical swine fever virus and foot and mouth disease virus,it significantly increased the antibody titers.RDS-treatment showed an excellent resistance to enhance T-SOD activity.Values of Blood routine and blood biochemistry showed no toxicity.These results suggested that RDS could be considered as an adjuvant to enhance immune responses to vaccines,as well as dietary additives for animals to enhance humoral and cellular immunity.
