Viperin?virus inhibitory protein,endoplasmic reticulum associated,interferon-inducible?is a member of ISGs which could act as effector molecules in immune response.Viperin could be divided into three parts,N-terminal domain,SAM domain and C-terminal domain.It is clear that viperin exhibitsability against various viruses through a broad array of methods.Duck and goose Viperin sequences are amplified and analysed.The result shows that sequence of duck Viperin includes 1092 nucleotides which codes 363 amino acids and partial sequence of goose Viperin includes 879 nucleotides which codes 292 amino acids.Analysis of amino acidsof duck and goose Viperin reveals that these two sequences are in highly similarity,exceptlack of N-terminalof goose Viperin.Duck Viperin sharesthe most closeevolution relationship with goose Viperin through the phylogenetic tree.Based on the sequences that have been obtained,real-timequantitative PCR method is used to explore the immunological properties of duck and goose Viperin.The result shows that DHAV,DPV,NDV and DTMUV could induce duck Viperin mRNA expression in DEFand DTMUV,GPV,H9N2,ODN 2006,LPS,R848 and Poly I:Ccould induce goose Viperin mRNA expression in goose PBMCs.Then,pcDNA3.1?+?-duck Viperin and pEGFP-C2-duck Viperin eukaryotic expression plasmidsare constructed.The result shows that duck Viperin weighs 42 kDa and only locates in the cytoplasmid of DEF and BHK21 cell.To explore if duck Viperin could inhibit replication of DTMUV in vitro,DEF are transfected with pcDNA3.1?+?-duck Viperin plasmids and pcDNA3.1?+?plasmids and then infected with DTMUV in 12 h post transfection.The detection of genomic copy number,TCID50 and expression of the fluorescence of DTMUVindicates that duck Viperin could inhibitreplication of DTMUV.In summary,this study focuses on cloning goose and duck Viperin,analying the similarity and immunological propertiesof two sequences and exploring the posibility of duck Viperin against DTMUV.
