| Chinese dominant traditional industry “Silk Industry” has a long history in the world,and mulberry trees play an indispensable role in silkworm food.Ge Lu mulberries are mainly distributed in many provinces,such as Shanxi.It is the most representative mulberry population in the Loess Plateau and is the important part of mulberry germplasm resource in China.Correlation analysis of agronomic traits and molecular markers of Ge Lu mulberry germplasm resources to find the phenotypic correlation with superior genes and explain the genetic variation of mulberry phenotypic traits from the molecular level.Which can provide the basis for cloning and excavation of important functional genes in mulberry germplasm resources and mulberry genetic breeding.In this paper,the agronomic traits of 98 Ge Lu mulberry varieties in Loess Plateau were identified in the field.Then we analyzed the genetic diversity and population structure of the participating germplasm.Finally,the correlation between agronomic traits and molecular markers was analyzed,and then we obtained the molecular marker loci that affecting agronomic traits.The research results are as follows:1.The data of 34 agronomic characters from 98 participating germplasm were analyzed by data processing software SPSS17.0.The results showed that there were significant differences among different agronomic traits,and the coefficient of variation was over 30% in total numbers of branchs per plant,leaf yield per plant(autumn)and the leaf numbers per kilogram(autumn).While the maximum variation coefficient of leaf yield per plant(autumn)was 45.44%.In addition,there is a high degree of diversity in leaf base,leaf color,leaf luster and leaf tip.The results showed that there was a rich genetic diversity in Ge Lu mulberry,which laid a foundation for further correlation analysis.2.We analyzed the genetic diversity and population structure of 98 accessions by using 11 ISSR primers.A total of 119 clear bands were amplified,and there were 97 polymorphic bands with a polymorphic ratio of 81.5%.The number of bands amplified by different primers was 9 ~ 13,and the average number of bands amplified by each primer was 11,and the average number of polymorphic bands was 9.Through the analysis of genetic diversity,the average number of alleles observed at each locus was 1.8235,the number of effective alleles was 1.5243,the Nei’s genetic diversity index was 0.3023,and the Shannon’s information index was 0.4480.The results showed that the ISSR of the 98 germplasms had a large variation,and there was a rich genetic diversity and high polymorphism.98 gerusanne germplasm were divided into 3 subgroups by structure group analysis,the results of UPGMA was consistent with the viewpoint of Xu Yongxin that “Qinshui county,yangcheng county,Shanxi Province is the original place of Ge Lu mulberry.3.The comprehensive analysis of phenotypic trait data,polymorphic marker locus data,population structure data(Q value)and kinship matrix(K)of 98 reference germplasms by software TASSEL2.1 and analyzed the association between molecular markers and agronomic traits by applying mixed linearity the model(MLM).There are 17 P sites associated with 14 agronomic traits at P<0.01 were detected among 119 ISSR sites,and the explanatory rate of phenotypic variation was 7.03%~12.27%.The whole leaf shape associated with the loci29 locus was the lowest interpretation rate of phenotypic variation and the loci119 locus(autumn)has the highest interpretation rate of phenotypic variation is the leaf yield per plant.There are two loci associated with the whole leaf shape,lenticels,and winter bud color,and three loci in the leaf yield per plant.In this paper,the results of agronomic traits association analysis and molecular markers of Ge Lu mulberry germplasm have provided rich data for mulberry inheritance and breeding,and have a certain guidance and reference for mulberry important functional gene mining and cultivate genetic improvement. |
