Davidia involucrata originates from ancient times and is a kind of precious plant in China,which belongs to the national first-class protection plant because of its endangered and extinct status.It is conducive for the protection,ecological and economic value development of D.involucrata to carry out the molecular biology research of this plant.However,the researches on D.involucrata mainly focus on its physiology and community structure,and rare on the molecular.Therefore,based on 9 gene fragments,this study used PCR direct sequencing method to sequence the D.involucrata materials in 18 counties of Sichuan Province.After collating and alignment these 160 sequences,analysis genetic relationship of D.involucrata in 18 counties of Sichuan Province were carried out.The main results are as follows:(1)D.involucrata in Baoxing,Dujiangyan,Hongya and Hanyuan’s were grouped in a small branch of the seven fragments:ITS,matK,rbcL,ndhF,trnS-trnG,trnL-trnF,rpl32-trnL,which indicates that the relationship between them is closely.D.involucrata in Ebian and the 4 were mostly in a small branch of the seven fragments.This indicates that there is a slight evolutionary difference between the two,but the relationship between them is relatively closely.(2)D.involucrata in Bifengxia and Qionglai were grouped in a small branch of the seven fragments:ITS,matK,rbcL,ndhF,trnS-trnG,trnL-trnF,rpl32-trnL.This indicates that the relationship between them is closely.(3)D.involucrata in Muchuan and Tianquan were grouped in a small branch of the matK,rbcL,and ndhF gene fragments,and among the remaining 4 gene fragments:ITS,trnS-trnG,trnL-trnF,and rpl32-trnL,were grouped in one large branch.This indicates that there is a slight evolutionary difference between the two,but the relationship between them is relatively close to that of D.involucrate in the other 16 origins.(4)D.involucrata in Pingshan and Meiguo were grouped in a small branch of the rbcL,ndhF and trnL-trnF gene fragments,and among the remaining 4 gene fragments:matK,ITS,trnS-trnG,and rpl32-trnL,were grouped in one large branch.This indicates that there is a slight evolutionary difference between the two,but the relationship between them is relatively close to that of D.involucrate in the other 16 origins.(5)D.involucrata in Gongxian and Beichuan were grouped in a small branch of the matK,rbcL and ndhF gene fragments,and among the remaining 4 gene fragments:ITS,trnS-trnG,trnL-trnF and rpl32-trnL,were grouped in one large branch.This indicates that the relationship between them is relatively close to that of the other originated D.involucrata.(6)D.involucrata Junlian and Leibo were grouped in a small branch of the matK,rbcL and ndhF gene fragments,and among the 3 gene fragments:ITS,trnS-trnG and rp132-trnL,were grouped in one large branch.They were not grouped in one large branch of the trnL-trnF gene fragments.This indicates that the relationship between them need further confirmed.(7)The trnL-trnF gene fragment was not detected in the D.involucrata from Dayi,but among the other 6 gene fragments used for cluster analysis,they were clustered together with D.involucrata in Bifengxia and Qionglai.This indicates that the D.involucrata in Dayi is relatively close to the D.involucrata in Bifengxia and Qionglai.(8)D.involucrata in Pingwu and Yingjin were grouped in a small or large branch of the matK,rbcL and gene fragments.D.involucrata in Yingjin was clustered as a single branch of the matK gene fragment.D.involucrata in Pingwu was clustered as a single branch of the rbcL gene fragment.D.involucrata in Yingjin and Junlian were in a small branch of the trnL-trnF gene fragments,while the D.involucrata in Pingwu was not detected the trnL-trnF gene fragment.In summary,this study explored Davidia involucrata relationships origination from eighteen counties in Sichuan Province,which guided for genetic research of Davidia involucrate.DNA sequences combination with morphological characteristics and cytogenetic examinations,as well as in situ conservation,increase gemination,etc.which may enhance researches of genetic resources,furthermore,protect Davidia involucrate.
