Preparation,Quality Evaluation And Preparation Of Clorsulon Liposome Study On Pharmacodynamics Of Fascioliasis Hepatica

Fascioliasis in sheep is a disease of great importance in veterinary medicine.It's all known that clorsulon has a positive effect on fascioliasis hepatica,unfortunately it is unstable and has a distinct toxic and side effect.As the reports,the liposomes of clorsulon can improve the stability and achieve a targeted effect,which is more conducive to the treatment of sheep fascioliasis hepatica.In this study,clorsulon was prepared into liposomes for characterization test,and subsequently observed the therapeutic effect in the infected sheep.In this experiment,the rotation-evaporation method was used to prepare chlorsulon liposomes.The encapsulation rate of liposomes was determined by HPLC which was used as the evaluation index for orthogonal test to screen the best prescription process.Finally,the therapeutic effect of liposome in sheep with fascioliasis was identified.The results showed that the standard curve was y=18675x±8514.4,R~2=0.9999,the control within-day precision relative standard deviation(RSD)was 0.87%~1.21%,daytime precision RSD was 0.92%~1.15%,average recovery percent was 99.69±0.36%,RSD was 0.46%.Best prescription preparation process for the quality as followed:phospholipid/cholesterol ratio was 2:1,medicine/fat was more than 1:2,ultrasonic 5 min,hydration in 37?,the income of the liposome envelopment rate reached 89.46±0.84%,respectively.Liposomes were round or oval,particle size were mainly distributed in the range of 400 nm to 500 nm,the average medicine content were 22.28±0.14%.The period validity could up to three months at 5?.Within 24h,in vitro the drug could release completely,and the phospholipids oxidation index was less than 0.2.The results of efficacy test showed that all the groups had an effective anthelmintic treatment which could restore the indexes and parameters of sheep liver function to a certain extent,and could improve the healthy of sheep flocks.The results showed that the liposomes in a suitable shape and have an uniform particle size distribution.This method was feasible and beneficial to accurately determine the encapsulation efficiency and medicine contents,and could meet the requirements of clinical medication.Moreover,sheep were intraperitoneally injected with liposome suspension(5 mg/kg),and the drug delivery group was continuously given for 3 days,2 time/d,which showed it could not only reduce the cost but also achieve a better therapeutic effect.
