Tiger grouper(Epinephelus fuscoguttatus)and hybrid grouper(E.fuscoguttatus ? × E.polyphekadion?)are fish of the genus grouper,which are important cultured species on the southeast coast of China.Hybrid grouper is a hybrid of brown spotted grouper(E.fuscoguttatus ?,also known as tiger grouper,brown spotted grouper)and E.polyphekadion ?(also known as Camouflage grouper).Studies at home and abroad have shown that the hybrid grouper has better growth traits than the parent,and is a hybrid with very market acceptance and suitable for industrial farming.The existing research on the hybrid grouper mainly focuses on apparent traits such as growth or physiological and biochemical surface,but the research on the molecular mechanism behind the growth has not been reported.In order to investigate the molecular mechanism behind the growth differences between brown spotted grouper and hybrid grouper,the brain,liver and muscle tissues of tiger grouper and hybrid grouper were sequenced and compared.The results obtained are as follows:1.7 581 722 449,6 330 804 586,8 175 941 387,6 343 265 141,6 229 084 451,7 197 785 696 bp clean reads were obtained by transcriptome sequencing in six libraries of brain,liver and muscle of hybrid grouper and tiger grouper,respectively.2.Assembly of 54 610 unigenes with a GC content of 46.666 5%,an N50 of 2 718 bp,an average length of 1 357 bp,and a total data volume of 74 119 587 bp.The assembled 54610 unigenes were aligned to the Nr,Swissprot,KOG and KEGG database functions,with a total of 28 832 unigenes annotated to the Nr,Swissprot,KOG and KEGG databases.3.Comparative analysis of brain tissue revealed 6 437 differentially expressed genes,further analysis found that growth hormone,growth hormone releasing hormone precursor,prolactin,prolactin precursor,arginine oxytocin and other growth-related genes in the hybrid stone The spotted fish is significantly up-regulated,which is consistent with the faster growing traits of the hybrid grouper.Comparative analysis of liver tissue revealed 7 380 differentially expressed genes,and found that phosphatidylinositol 4-phosphate 5-kinase 1? type,protein phosphatase 1 regulatory subunit 3C,liver-fatty acid binding protein,starch binding domain protein 1,Mid1-interacting protein 1 were up-regulated in the hybrid grouper and also in line with the faster growth of the hybrid grouper.Comparative analysis of muscle tissue revealed 6 417 differentially expressed genes,in which the genes encoding the muscle development such as the paired box protein Pax-3,creatine kinase subtype,myocardin-like and MyoD were significantly up-regulated in the hybrid grouper.,which consistent with the hybrid grouper growing faster traits.The differentially expressed genes were aligned into the KEGG database.A total of 7452 differentially expressed genes were aligned into 217 pathways.Among them,neuroactive ligand-receptor interaction,endocytosis,MAPK signaling pathway,focal adhesion and calcium signaling pathway are pathways for enriching more genes,among which MAPK signaling pathway,calcium signaling pathway,PPAR signaling pathway are closely related to growth regulation,which involved in the regulation of the differential growth of hybrid grouper and tiger grouper.4.The platelet-derived growth factor-like receptor,fatty acid binding protein and MID1-interacting protein 1 gene were cloned,and the bioinformatics analysis of the sequences of these three genes was carried out.Finally,the tissue distribution of PDGFRL,FABP and MID1 IP genes was found.These three genes are expressed in various tissues.The expression of PDGFRL in the heart is significantly higher than that in other tissues.The expression of FABP in the liver is significantly higher than that in other tissues.The expression of MID1 IP in the heart,muscle and intestine is significantly higher than other tissues..In summary,the transcriptome data of brain,liver and muscle of hybrid grouper and tiger grouper were obtained,and a number of growth-related differentially expressed genes were screened.These genes were differentially expressed in brain,liver and muscle.It may be an important factor in the difference in growth of two groupers.At the same time,the cloning and characterization of PDGFRL,FABP and MID1 IP gene sequences of hybrid grouper were cloned,which laid a foundation for the study of functional verification and regulation mechanism of hybrid grouper.
