Antheraea pernyi is a valuable and developable economic insect because it can produce silk and be eaten.But Beauveria bassiana can kill A.pernyi,which causes serious losses in sericulture.In this study,we used the molecular biology methods to identify the pathogen of batryticatus,which have the same symptom.The microbial diversity of indoor and outdoor in the oak silkworm's habitats was analyzed by Illumina Miseq high-throughput sequencing technology.The results will provide new molecular identification methods and theoretical basis for sericulture pruduction.The main findings are as follows:1.The ITS and specific primers were screened to identify the pathogen of batryticatus by PCR technology.The sequence homology was different between the collected samples in different years and artificially inoculated samples.2.We tested different soil microbial DNA extraction methods and found that low-temperature freeze extraction method is suitable for identification the batryticatus pathogen in the soils.The distribution results of pathogen showed that the detection rate of fungal and B.bassiana is 83.33% and 46.08% among 204 samples from 68 collection sites.3.The Illumina Miseq technique was used to determine the microbial diversity of indoor and outdoor in the oak silkworm's habitats.A total of 14 doors,37 classes,93 orders,201 families,346 genera fungi and 37 doors,99 classes,128 orders,260 families,477 genera bacteria were detected from the indoor and outdoor habitats.
