with a long history of rice farming.The rice-fish farming system in Qingtian District was approved by the FAO as one of the first Globally Important Agricultural Heritage Systems(GIAHS)in 2005.After long-term evolution in the rice field environment,Cyprinus Carpio var Color has a variety of unique structures.Among them,some scholars have found that the scales of the fish have soft scales and hard scales.Cyprinus Carpio var Color in Qingtian have soft scales,while in Jinhua are dominated by hard scales.This paper selected Cyprinus Carpio var Color from Qingtian and Jinhua in Zhejiang Province as research materials and compared muscle nutrients,tissue water content and aquaporin 1 tissue expression levels in them,designed to find other differences in Qingtian and Jinhua Cyprinus Carpio var Color,hoping to provide evidence and reference for further revealing the reasons for the differences between the two scales.The specific research contents are as follows: 1.Analysis of muscle nutrient composition of Cyprinus carpio var color in Qingtian and Jinhua.Nutrient composition of Cyprinus carpio var color in Qingtian and Jinhua were analyzed by generic methods.The results showed that the content of water and crude fat(fresh weight)were significantly higher in Qingtian Cyprinus carpio var color;The muscles of them all contained 17 amino acids.The percentage of amino acids in Jinhua Cyprinus carpio var color were higher than Qingtian Cyprinus carpio var color but Glycine and Cystine;Based on the criteria of amino acid score(AAS),the top two limiting amino acids in Qingtian Cyprinus carpio var color were Valine and Methionine+Cysteine,and the top two limiting amino acids in Jinhua Cyprinus carpio var color were Methionine+Cysteine and Valine;16 fatty acids were detected in all fish muscles.The C16:1,C18:1n9c,C18:3n3 and monounsaturated fatty acid were higher in Qingtian Cyprinus carpio var color;and the C18:0 and C20:4n6 were higher in Jinhua Cyprinus carpio var color.The content of Zn and Cu in Qingtian Cyprinus carpio var color were significantly higher than Jinhua Cyprinus carpio var color,and the content of Ca,P and Mg in Jinhua Cyprinus carpio var color were significantly higher than Qingtian Cyprinus carpio var color..2.Analysis of muscle nutrient composition of Cyprinus carpio var color under different aquacultural settingsNutrient composition of Cyprinus carpio var color under different aquacultural settings were analyzed by generic methods.In the paddy field culture mode,the crude fat content of the muscles of Cyprinus carpio var color was significantly higher than that of the pond culture mode.17 common amino acids were measured in the muscles of the 6 treatment groups.The histidine content of Cyprinus carpio var color in group F was significantly lower than that of other groups.The MUFA content of muscles in group E was significantly higher than that in group F,and the content of C16:0 was significantly lower than that of other groups.The content of C20:4n6 and docosahexaenoic acid C22:6n3 in group F was significantly higher than other groups.Calcium content in group A and group F was significantly higher than other groups.The content of magnesium in group F was significantly higher than that in group C,and the content of zinc and selenium in group B was significantly higher than that in group F.3.Comparative study on tissue water content between Cyprinus carpio var color in Qingtian and Jinhua.Tissue water content between Cyprinus carpio var color in Qingtian and Jinhua were analyzed by generic methods.The water composition of the muscles,scales and gill tissues of the fish in the two regions was compared by LF-NMR.The results showed that the water content of gill and muscle of Cyprinus carpio var color in Qingtian was significantly higher than that in Jinhua.The water content in scales of Cyprinus carpio var color in Jinhua was significantly higher than that in Qingtian.The distribution and content of water in gills,muscles and scales in Qingtian and Jinhua Cyprinus carpio var color showed great differences.The proportion of muscle-bound water in Qingtian Cyprinus carpio var color is higher than that of Jinhua.4.Molecular cloning and expression analysis of aquaporin 1 in Qingtian and Jinhuatian Cyprinus carpio var color.The cDNA full-length sequences of AQP1 in Qingtian and Jinhua Cyprinus carpio var color were obtained by RACE technique respectively,and the expression of AQP1 in their tissues was analyzed.The full-length of AQP1 cDNA sequence in Qingtian Cyprinus carpio var color is 1197 bp,of which 5’UTR is 66 bp,3’UTR is 351 bp,and the ORF is 780 bp,encoding a total of 259 amino acids.Jinhua Cyprinus carpio var color AQP1 is 1221 bp in length,of which 5’UTR is 66 bp,3’UTR is 375 bp,ORF is 780 bp,encoding 259 amino acids.The theoretical isoelectric point of Qingtian Cyprinus carpio var color AQP1 is 5.77,and the molecular weight of the protein is 27.2kDa.The theoretical isoelectric point of Jinhua Cyprinus carpio var color is 6.13,and the molecular weight of the protein is 27.3kDa.AQP1 expression level in tissues indicated that AQP1 had the
