| (Peperomia tetraphylla(Forst.F.)Hook.et Atm.)has a thick green leaf,such as jade-like blue,and the flowers are fresh and pleasant,belonging to the "small fresh" of the foliage plants.In this study,the red-breasted green sterile tube seedlings were used as explants,and the red-body chloroplast cell suspension was carried out on different concentrations of exogenous hormones,medium concentration,carbon source,shaking speed and continuous culture time.Cultivate research.At the same time,the air culture environment and callus cytology of the test tube seedlings were studied.The survival rate of the test tube seedlings was improved by transplanting and domestication,which provided a scientific basis for the later(Peperomia tetraphylla(Forst.F.)Hook.et Atm.)engineering and production application.The main findings of this test are as follows:1.(Peperomia tetraphylla(Forst.F.)Hook.et Atm.)embryo cultured cell suspension culture(1)The optimal embryogenic callus culture medium of(Peperomia tetraphylla(Forst.F.)Hook.et Atm.),6g/L agar,30g/L sucrose,1.0 mg/L 6-BA,1.0 mg/L 2,4-D were added to MS medium.Exogenous hormone,induction rate of 71.1%,formation of loose green callus;embryonic cell formation and proliferation medium formula:MS liquid medium added 30g/L sucrose,0.5 mg/L NAA and 1.0 mg/L6-BA,the induction rate of embryogenic cell mass was 66.12%.(2)The tissue cell suspension culture process was initially inoculated with 0.4 g tissue cells in liquid medium,and the fresh weight of the cells was 2.1 g,and the proliferation rate was the largest.The addition of 30g/L sucrose was the best carbon source for the proliferation of(Peperomia tetraphylla(Forst.F.)Hook.et Atm.),and the fresh weight of the cells was 1.56g.After continuous culture for 42 days,the cell proliferation was the largest,the cell mass grew well,and the fresh weight was 1.67 g.The shaking speed of the shaker is 110 r/min,and the cell cluster has the best proliferation effect.(3)The optimal medium concentration of green cell cluster differentiation and plant regeneration process was 1/2 MS.The best differentiation medium for somatic embryo differentiation and plant regeneration was MS+1.0m/L6-BA+0.3 mg/L IBA+30g/L sucrose+6g/L agar.(4)The process of green somatic embryogenesis of(Peperomia tetraphylla(Forst.F.)Hook.et Atm.)It experiences a small spherical embryo,an elliptical embryo,a heart-shaped embryo,a cotyledon,and a mature embryo stage.2.Study on gas rooting inhibition and domestication techniques of(Peperomia tetraphylla(Forst.F.)Hook.et Atm.)(1)The greening root inhibition process of red bean peas was carried out on a medium supplemented with 30 g sucrose,5 g agar,0.1 mg/L NAA,0.5 mg/L 6-BA at a concentration of 1/2 MS,and the number of aerial roots was the least.6-BA and NAA played a major role in the inhibition of(Peperomia tetraphylla(Forst.F.)Hook.et Atm.)(2)NAA,IAA,I-BA three kinds of auxin,NAA inhibited air rooting effect best,I-BA effect was second,IAA effect was the worst,and the amount of gas rooting was the least on medium with 0.3 mg/L NAA.(3)The concentration of the supporting substance on the red-edged green bean medium was 1/4 MS,which inhibited the formation of aerial roots.2g of perlite was added to the medium to form the least amount of aerial roots.The plants were continuously cultured on a medium of 2 g perlite+0.3 mg/L NAA+0.5 mg/L 6-BA+30 g/L sucrose+5 g agar for 35 to 42 days,and the number of aerial roots was the least.(4)The process of embryogenic callus induced by red-edged green leaves,and some callus dedifferentiated to form a large number of aerial roots.On the MS medium supplemented with 2,4-D 0.5 mg/L+6-BA 0.5 mg/L,yellow-green embryogenic callus was formed to differentiate into somatic embryos,which were then differentiated to form intact plants,which inhibited the formation of aerial roots.(5)The red-edged watercress green transplanted water for 6d has the best effect,and the less the number of gas-rooted roots,the higher the green survival rate of the red-edged watercress.With vermiculite:perlite:peat=1:1:2 for the transplanting of the test tube seedlings,(Peperomia tetraphylla(Forst.F.)Hook-et Atm.)grows robustly. |
