| The F1 generation fattening castratied cattle of Japanese black cattle x Qinchuan cattle were fed with Pasteur biological feed,the body weight was measured regularly during the feeding period,and the post-mortem production was measured and evaluated after slaughter.The technology real-time quantitative PCR(qRT-PCR)was used to determine the expression of TG,PPARa and PPARβgene in the experimental group and the control group,and to analyze the molecular mechanism of the effect of feeding Pasteur biofeed on the fat deposition of beef cattle.Experiment 1:Selecting 40 F1 generation fattening cattle of Japanese black cattle and Qinchuan cattle with similar age(28-29 months old),randomly divided into control group and experimental group,adding 0%and 8%of feeding standards respectively.The feeding period is 5 months,and the body weight is measured once a month.At the end of the feeding trial,14 cattle were randomly selected from each group for slaughter,and the 12-18 intercostal eye muscle area was measured and the marbling was scored.The results showed that the average daily weight gain of the experimental group was slightly higher than that of the control group(P>0.05),and the eye-muscle area and marbling score were significantly higher than the control group(P<0.01).Experiment 2:The total RNA of ocular muscle tissue(meat between the 12-13 ribs of each group carcass)was extracted by Trizol method(14 cattle in each group)and qRT-PCR was performed to detect the expression levels of TG,PPARa and PPARβ genes.The results showed that the expression levels of TG and PPARa in the experimental group were significantly higher than those in the control group(P<0.05),and the expression of PPARβ gene was higher than that in the control group,but it did not reach significant level(P>0.05).Experiment 3:Correlation analysis was performed on the data of the first and second experiments.The results showed that there was a significant correlation between the marbling traits of beef cattle and the expression of gene TG,PPARa and PPARp(P<0.05).The above experiments showed that the addition of 8%Pasteur biological feed in the F1 generation fattening cattle diet of Japanese black cattle x Qinchuan cattle could improve the marbling score and increase the eye muscle area,and significantly promote the expression of TG and PPARa genes.This can provide a theoretical basis for cultivating high quality beef cattle and increase the production. |
