It is great significance to elucidate the molecular mechanism of rice heat resistance and accelerate the breeding of new varieties of heat-resistant rice to ensure China's food security.So,this study use near-isogenic line of heat resistant rice pure line XN0437 T and heat sensitive rice pure line XN0437 S for material and use artificial climate box to conduct night temperature treatment at early milky stage.iTRAQ protein labeling technology is used to study the heat resistant rice pure line XN0437 T and heat sensitive rice pure XN0437 S of caryopsis of the proteome response patterns mode and proteome difference in response to high nighttime temperature stress at early milky stage.The main results are as follows.(1)According to the results of mass spectrometry,the total number of detected peptides was 65,578 compared with Uniport database,and 14,497 kinds of peptides were identified by filtering and screening with high reliability.A total of 3,130 high quality proteins were identified and quantitatively identified under the screening criteria of FDR(false discover rate)1%.In the GO analysis of these identified proteins,it was found that they were involved in 21 biochemical processes,7 molecular functions and 1 cell composition in the caryopsis cell.KEGG pathway analysis showed that these proteins were concentrated in the endoplasmic reticulum protein processing signaling pathway with the highest significance.(2)Compared with the control rice material growing under the appropriate temperature conditions,there were 55 up-regulated proteins(FC > 1.5)and 16 down-regulated proteins(FC < 0.667)in the kernels of heat-resistant rice pure line XN0437 T.There were 32 up-regulated proteins and 15 down-regulated proteins in the caryopses of the heat-sensitive rice pure line XN0437 S.Among them,16 proteins were up-regulated 1.5 times and 8 proteins were down-regulated in both heat-resistant and heat-sensitive rice.(3)Heat resistant and heat sensitive rice protein function annotation further show that there are seven proteins involved in signal transduction,there are six protein involved in material transportation,there are three proteins involved in gene transcription,there are two proteins involved in REDOX,there are seven proteins involved in heat shock response,there are four proteins involved in photosynthesis,two participate in energy metabolism and fourteen material synthesis,there are forty-nine protein function is unknown.(4)The function and expression pattern of differentially expressed proteins were analyzed and it was found that heat shock protein showed the same expression pattern in the two rice lines,which were up-regulated.This is also a short-term stress response of rice under high temperature stress.As representative zinc finger proteins,Q5YLY5 and Q10N88 are closely related to transcriptional signal transduction and their expression patterns are conducive to improving the heat resistance of rice.However,in two pure rice lines,it was found that both P17048 and Q6ZIX4 showed the same down-regulation expression,which also indicated that high temperature at night had an impact on the quality of rice.
