| The aim of this study was to explore the effects of age on the carcass performance and meat quality traits of Huainan Partridge chicken,as well as the expression patterns of fat and meat candidate genes,and provide evidence for screening candidate genes for meat quality traits.The experiment was conducted on Huainan Partridge chicken,and 80 birds were raised from the same batch,with half male and female.20 birds were randomly selected at 90,120,150 and 180 days of age,half for carcass performance,meat quality,blood lipid and candidate gene expression analysis.The results are as follows:I.Carcass performance and muscle quality in Huainan Partridge chickenThe live weight,carcass weight,carcass rate,semi-eviserate and eviserate of Huainan Partridge chicken were highest at 180 days,the male slaughter rate was 86.06%-90.48%,and the pectoral muscle rate was 14.35%-18.22%,the leg muscle rate was 31.84%-35.34%,the abdominal fat percentage was 0.09%-0.31%.At 180 days of age,the shear force of pectoral muscle and leg muscle was 20.95N and 28.79N.The IMF content in pectoral muscle and leg muscle was 2.31-4.60%and 4.08-8.76%,and the IMP content in pectoral muscle and leg muscle was 1.68-2.46%and 1.39-2.38%;The slaughter rate of hens was 80.75%-90.84%,the chest muscle rate was 15.86%-17.31%,and the leg muscle rate was 28.75%-31.17%.The abdominal fat percentage was 0.44%-4.24%,the shear force of pectoral muscle and leg muscle at 180 days of age was 21.09N and 27.88N,the IMF content in pectoral muscle and leg muscle was 2.75-7.25%and 3.02-6.70%,and the IMP content in pectoral muscle and leg muscle was 1.53-2.32%and 1.70-2.27%.2.Analysis of blood lipid index and its correlation with intramuscular fat contentThe concentrations of TC and VLDL in serum were the highest at 180 days,the highest values of TC and VLDL were 19.36 mmol/L and 10.83 mmol/L,and the highest values of TC and VLDL in females were 24.58 mmol/L and 15.36 mmol/L.The concentration of FFA and LDL in male was the highest at 120 days,and the concentration of TG was the highest at 180 days.The FFA,TG and LDL of females were the highest at 180 days old.IMF levels of chest and leg muscles were positively correlated with HDL concentrations(r=0.3822,P<0.05;r=0.4236,P<0.01).In the female,there was a significant negative correlation between IMF content and TG concentration(r=-0.3639,P<0.05).3.Gene expression and its correlation with intramuscular fat and inosinic acid contentThe expression levels of liver,breast muscle and leg muscle of male FASN gene were highest at 180,120 and 90 days old respectively.The expression levels of liver,breast muscle and leg muscle of LPL gene were highest at 150,120 and 180 days old respectively;liver and breast muscle of AMPD1 gene The expression levels of the leg muscles were highest at 90,90,and 150 days,respectively;the expression levels of the ADSL gene in the liver,chest muscles,and leg muscles were highest at 180,120,and 150 days,respectively.The expression levels of liver,breast muscle and leg muscle of female FASN gene were highest at 150,180 and 90 days old respectively.The expression levels of liver,breast muscle and leg muscle of LPL gene were highest at 150,120 and 180 days old respectively;liver and breast muscle of AMPD1 gene The expression levels of the leg muscles were highest at 90,90,and 150 days,respectively;the expression levels of the ADSL gene in the liver,chest muscles,and leg muscles were highest at 150,180,and 90 days,respectively.There was a significant positive correlation between IMF content and FASN gene,FASN gene and LPL gene expression in pectoral muscle of male(r=0.5343,r=0.4017,r=0.5385,P<0).05).There was a significant positive correlation between IMF content and FASN gene expression in pectoral muscle and LPL expression in leg muscle of female(r=0.6253,P<0).05);In the male,there was a significant positive correlation between chest IMP content and AMPD1 expression(r=0.4142,P<0).05),there was a significant positive correlation between IMP and AMPD1 expression(r=0.5343,P<0).(05)was significantly positively correlated with ADSL(r=0.6754,P<0).01).In the female,there was a significant positive correlation between IMP content and AMPD1 expression(r=0.8049,P<0).01),IMP was negatively correlated with AMPD1 and ADSL expression in the liver(P<0).05).In summary,the age has a significant effect on the growth and meat quality of Huainan Mahuang chicken,which can be selected by the blood lipid index HDL and TG.FASN and LPL regulate intramuscular fat content,ADSL and AMPD1 regulate muscle inosinic acid content,and these genes are important genes affecting the meat quality traits of Huainan Mahuang chicken. |
