The Ujumqin sheep is an excellent meat population.The Ujumqin sheep meat is tender and juicy,with a small astringency,rich in a variety of minerals,a lot of fat and a dense but fat.Intramuscular fat(IMF)is an important factor affecting the quality of meat.Therefore,this study explored the differences in meat quality and the regulation of intramuscular fat in different parts of the skeletal muscles in different parts of the 8-month-old Ujumqin sheep.To lay a better foundation for improving the quality of Ujumqin mutton.Firstly,the crude fat and fatty acid of the longissimus dorsi,biceps femoris and triceps muscles of Ujumqin sheep were measured.Then the IDA technique and SWATH technique were used to identify the total muscles of the biceps femoris and the longissimus dorsi muscle.Protein and differential protein,GO enrichment analysis and KEGG pathway analysis of total protein and differential protein,respectively,found that pathways and proteins may be involved in intramuscular fat metabolism;finally selected differential proteins for Western blot quantitative verification,the main findings are as follows:(1)Through the detection of crude fat and fatty acid in three muscles of Ujumqin sheep aged 8 months,it was found that the content of crude fat in the biceps femoris was significantly higher than that in other parts(P<0.01).A total of 36 species were identified.The total content of fatty acid in the biceps femoris was significantly higher than other sites(P<0.01);C15:1,C16:0,C18:0,C18:2c6 content was significantly higher in the biceps femoris than other The site(P<0.01);the content of unsaturated fatty acids,medium-chain fatty acids,n-3,n-6 series fatty acids beneficial to the human body in the biceps femoris were higher than the two parts.(2)Identify 852 protein of the longissimus dorsi muscle of Ujumqin,777 protein of biceps femoris muscle,and 998 total proteins in two parts.GO enrichment analysis and KEGG pathway analysis of total protein It was found that 828 biological processes(BP),161 cell components(CC),214 molecular functions(MF)and 64 pathways were enriched.(3)Screening 87 different proteins of the longissimus dorsi and biceps femoris muscles by SWATH method,including 56 proteins with up-regulated biceps femoris,31 proteins with down-regulated with biceps femoris;in the GO enrichment analysis and KEGG pathway analysis of differential proteins,it was found that 126 biological processes(BP),65 cell components(CC),52 molecular(MF)were enriched.And 25 pathways,of which 1/3 are associated with fat and fatty acid metabolism.(4)Verification of the expression of differential protein ACADM in two parts muscle by Western blot showed that the expression of biceps femoris was higher than that of the longissimus dorsi muscle(P<0.01),which was consistent with the proteome results.(5)Through KEGG pathway analysis found that differences in protein and fat metabolism related important pathways of PPAR signaling pathways,and the pathways of FABP3y CPT1B,ACADM protein to build protein interaction network found that FABP3,CPT1B,ACADM protein may with PPAR pathway of PPARa,PPARγinteraction to adjust Ujumqin sheep intramuscular fat metabolism,more research and validation should be carried out in the level of cells.
