| Global warming has become an international issue that cannot be ignored.Elevated CO2 concentration is the main problem of climate warming.The increase in atmospheric CO2 concentration will affect human society and agroforestry ecosystems.The Climate Comission reported that CO2 has reached its highest level in history in recent years and will continue to rise.In this paper,three CO2 concentrations were set according to the current atmospheric CO2 concentration and the predicted future CO2 concentration as increasing trend,with 397μL/L as the control and 550μL/L and 750μL/L as the treatment groups.The CO2 concentration(397μL/L,550μL/L,750μL/L)wasdirectly applied to the growth of the Lymantriadispar eggs to the third instar larvae,and LdNPV(397μL/L LC20=1.1×10PIB/μL)infected the third instar L.dispar larvae.After 24 hours,the L.dispar larvae were reared under the condition of 397μL/L CO2.The growth period of 3rd-4th instar L.dispar,the cumulative growth rate of body weight and cumulative mortality were measured.The activities of protective enzymes and detoxification enzymes in Ldispar were determined,and median lethal concentration(LC50)of LdNPV was determined under different CO2 concentrations.High-through put techniques were used to construct transcriptomes of third instar larvae,LdNPV(LC20=1.1×10PIB/μL)treated-and normal larvae after feeding with different CO2 concentrations(397μL/L,550μL/L,750μL/L).The differential gene expressions were compared and analyzed,and the OBP family genes of Ldispar was screened for in situ hybridization tissue localization.The main results are as follows.1.The median lethal concentration(LC50)of 397μL/L and 550μL/L of LdNPV were 7.54×lO2PIB/μL,4.85×102PIB/μL and 3.03x102PIB/μL,respectively.The cumulative growth rates of body weight of the third instar larvae of L.dispar reared underdifferent CO2 concentrations(397,550and 750μL/L)were 81.27%,71.63%and 68.41%higher than those of the control group,respectively.The mortality of LdNPV infection increased with the increase of CO2 concentration and that of the 750μL/L treatment group was 27.09%.Increased CO2 concentration increased efficiency of the conversion of ingested food and efficiency of the conversion of digested food of the third instar larvae of.Ldispar to maintain growth and development,so that LdNPV in the worms will multiply and slowdown their growth and development,which may lead to LdNPV treatment under high CO2 treatment.L.dispar larvae have a long incubation period and a high mortality rate.2.The activities ofdetoxifying enzymes CarE and AChE in the third instar larvae of L.dispar increased with the increase of CO2 concentration,while the activities of ALP decreased with the increase of CO2 concentration.The activities of protective enzymes CAT decreased with the increase of CO2 concentration,while the activities of SOD increased with the increase of CO2 concentration.After inoculation with LdNPV,the activities of CarE,ALP,AChE and CAT were inhibited,while the activities of SOD were induced to increase with the increase of CO2 concentration.The activity of CarE,AChE,ALP and CAT in the third instar larvae of L.dispar decreased with the increase of CO2 concentration.The destruction of the L.dispar immune defense system may cause the population of L,dispar todecrease.3.The transcriptomes data base of the third L.ispar instar fed with different CO2 concentrations(397,550and 750μL/L)treated by LdNPV(LC20=1.1×10PIB/μL)and the control were constructed;80.65Gb data were obtained and Q30 base was over 92%.Total 33895 of Unigene were obtained.19286 genes were annotated in Nr protein bank,12189 genes were annotated in KOG protein bank,12694 genes were annotated in SwissProt protein bank,13986 genes were annotated in KEGG protein bank,4205 genes were annotated in GO protein bank,13776 genes were annotated in InterPro proteinbank.The differential expressed genes were concentrated and responded to a total of 32 genes in all treatments.Four of the 32 differential expressed genes belonged to the trypsin family.By fluorescent quantitative RT-PCR analysis,trypsin(CL922.Contig4_GM1)was used as an immune defense-related gene in each treatment group(550CK,750CK,397NPV,550NPV,750NPV).The median is down.4.LdOBP1 gene is more distributed in antennae of female L.dispar adults than that of male L.dispar adults.LdOBP2 gene was more distributed in male L.dispar antennae than in female L.dispar adult antennae.In the antennae of Ldispar male adults,the distribution of LdOBP1 gene was lower than that of LdOBP1 gene.On the contrary,the distribution of LdOBP1 gene was higher than that of LdOBP2 gene in the antennae of female L dispar adult. |
