Plant biodiversity is the foundation of forest sustainable development and breeding,the population diversity of plants depends on its population structure,and determines the population utilization potential and way.Mating system is one of the most important factors affecting plant population structure.Sinomanglietia glauca is a rare and endangered plant in Magnoliaceae,which is in the connecting position between Magnolia and Manglietia in evolution.It has a straight trunk shape,magnificent flowers and fruits,and a faint fragrance when flowering,so it has a good prospect of development and utilization.In view of the weakness of the prophase studies,we started from the reproductive ecology,and then carried out the biological research of Sinomanglietia glauca from the transcriptomics,so as to provide a basis for the breeding of S.glauca.The main results are as follows:1.There was self-incompatibility in S.glauca,which was late-acting self-incompatibility.The results showed that the Selfed fruits fell off after 30d post pollilation,and the crossed fruit experienced 140d post poliilation to normal maturity.Compared with the fruits of Sinomanglietia glauca,we found that the undeveloped seeds in mature fruits still keep the size of ovule,which was obviously different from normal seeds.In contrast,the seeds of self-abortion fruits had developed and were obviously larger than the ovules.2.Transcriptome analysis of self-crossing and cross-crossing responses to gene changesTranscriptome sequencing was performed on the pistils of different pollination treatments:selfing(S),outcrossing(C)and control(CK).A total of 69.23 Gb of raw data were obtained from 9 samples by Illumina RNA-seq second-generation sequencing.After quality control,the proportion of clean reads in each sample was over 95%,and the Q30 of each sample was more than 93.53%.9 samples were mixed(SGM)and sequenced by PacBio Sequel platform for the third generation.a total of 16.39 Gb of data and 213381 transcripts were obtained.After second generation data correction to remove redundancy,a total of 135040 Unigene were obtained.The 135040 Unigene obtained were aligned with seven major databases including NR,NT,Pfam,KOG,swiswis-prot,KEGG and GO.There were 119251(88.31%)Unigene annotated on at least one database,and 32063(23.74%)Unigene were annotated by 7 major databases at the same time.Through the analysis of the expression levels of the differentially expressed genes between S vs CK,C vs CK and S vs C,there were 8201 up-regulated genes and 5395 down-regulated genes in S vs CK,7699 up-regulated genes and 5639 down-regulated genes in C vs CK group,151 up-regulated genes and 208 down-regulated genes in S vs C group.The differentially expressed genes were analyzed by GO and KEGG enrichment.The differentially expressed genes of the three groups were mainly enriched in the biological process,and the Photosynthesis-antenna proteins and the Starch and sucrose metabolism pathways.
