The preservation of ram semen can significantly improve the utilization efficiency of semen,reduce the cost of raising livestock and raise the breeding efficiency.However,there are few reports on the effective preservation of ram semen.Although the structure of sperm is simple,in the preservation of semen,the plasma membrane of the sperm is susceptible to external environmental conditions,result in changes in the structure,physical and chemical properties of the plasma membrane,resulting in a significant decrease or loss of sperm motility.Sodium cromoglicate(SC),acetylsalicylic acid(ASA),sodium salicylate(SS)as a common cytoplasmic membrane protector in the treatment of human silicosis and rheumatic joints.It is widely used in inflammation,but no relevant research has been found in the process of semen preservation.In this study,using Dubo ram semen as treatment subject,the protective effects and effects of SC,ASA and SS on the sperm plasma membrane of sperm under 20? and 4? conditions were investigated,in order to save the ram semen effectively.The foundation is widely used in the production process.Results,After adding three plasma membrane protector at 20?,the acrosome integrity(AI)of the group add SS or ASA was significantly higher than that of the control group at 6 h(P<0.01);the membrane integrity(MI)of the group with SS and ASA was significantly higher than control at 72 h(P<0.01).The MI of sperm in group with ASA and SS was significantly higher than that of the control(P<0.01);the sperm motility of the ASA group was significantly higher than that of the control group at 96 h(P<0.01),and MI remained extremely significant.compared with the control group(P<0.01),The Al of the ASA and SS groups was significantly higher than that of the control group(P<0.05).After adding 20 mM,50 mM,100 mM ASA,the sperm motility of the group supplemented with 50 mM and 100 mM ASA was significantly higher than that of the control group at 48 h(P<0.01),and the ASA group was added with 100 mM.The MI was significantly higher than that of the control group(P<0.05).The Al of the 100 mM AS A group was also significantly higher than that of control(P<0.05).At 72 h,the AI of the 100 mM ASA group was significantly higher than that of the control group(P<0.01).After adding 10 mM,20 mM,50 mM SS,the sperm motility of the 50 mM SS group was significantly higher than that of the control group at 24 h(P<0.05);At 48 h,the sperm motility of the 10 mM and 20 mM SS groups was significantly higher than that of the control group(P<0.05);at 72 h,the AI of all the treatment groups was significantly higher than that of the control group(P<0.01).After adding three plasma membrane protector at 4?,At 6 h,the Al of AS A group was significantly higher than that of the control(P<0.05).At 48 h,the MI of the SC group and the ASA group was significantly higher than that of control(P<0.05),and the SS group was significantly higher than the control group(P<0.01);at 96 h and 144 h,the sperm motility and MI of the ASA treatment group was significantly higher than that of the control group(P<0.01):the MI and AI of SS were also significantly higher than the control group at 144 h(P<0.01).After adding 20 mM,50 mM,100 mM ASA,At 48 h,the AI of the 20 mM and 100 mM ASA group was significantly higher than that of the control group(P<0.05).At 96 h,the sperm motility of all the treatment groups was significantly higher than that of the control group(P<0.05);at 144 h,the MI of the 50 mM ASA group was significantly higher than the control group(P<0.05),the AI of the 100 mM ASA group was significantly higher than that of the control group(P<0.05).After adding 10 mM,20 mM,50 mM SS,the sperm motility of the 20 mM SS group was significantly higher than that of the control group at 96 h(P<0.01).The sperm motility of the 10 mM SS group was significantly higher than that of the control group(P<0.01),and the 50 mM SS group was significantly higher than the control group(P<0.05);AI with 50 mM SS group was significantly higher than the control group(P<0.01);after 144 h,20 mM SS group was added.The sperm motility and MI were significantly higher than the control group(P<0.01),and the sperm motility of the 10 mM and 50 mM SS groups was significantly higher than that of the control group(P<0.05);AI of the 20 mM and 50 mM SS groups was significantly higher than the control group(P<0.01).Conclusion:ASA and SS significantly improve sperm motility,MI and AI in the semen preservation at 20? and 4? in ram semen.
