Using potato 'Feuerui' variety as test material,tissue culture method was used to study the freshness of stem,leaf and root of potato virus-free seedlings with 0,10,15,20,30,50,100 and 125mmol/LNH4NO3.Weight,dry weight,plant height,number of leaves,number of branches,total nitrogen content and effects of sucrose,fructose,soluble total sugar,glucose and starch.0,0.01,0.02,0.03,0.05,0.1,0.5,1,2mmol/L ZnSO4,0,0.05,0.1,0.15,0.2,0.25,0.5,0.75,1mmol/L MnSO4 and 0,0.05,0.1 were studied.0.15,0.2,0.4,0.7,1,2mmol/L H3BO3 fresh weight,dry weight,plant height,number of leaves,branch number and sucrose,fructose,soluble total sugar,glucose in stems and leaves of potato virus-free seedlings And the effect of starch content.The main results are as follows:1.The fresh and dry weight,plant height,leaf number and branch number of stem and leaf and root of potato virus-free seedlings increased first and then decreased with the increase of NH4NO3 concentration.Fresh weight and dryness of stems and leaves The weight,plant height and leaf number were all optimal with 20mmol/L NH4NO3,and the total nitrogen content increased with the increase of NH4NO3 concentration.2.With the increase of NH4NO3 concentration,the contents of sucrose,fructose,total soluble sugar,glucose and starch in the leaves of potato virus-free seedlings increased first and then decreased,and the treatment with 20mmol/L NH4NO3 was the highest.3.With the increase of the concentration of ZnSO4,MnSO4 and H3BO3 in the medium,the fresh weight,dry weight,plant height,number of leaves and number of branches of potato virus-free seedlings increased first and then decreased.The fresh and dry weight of stem and leaf treated with ZnSO4 was the highest at O.lmmol/L.The fresh and dry weight of stems and leaves under 0.5mmol/LMnSO4 treatment was the highest.The fresh,dry weight and plant height of stems and leaves treated with 0.4mmol/L H3BO3 were the highest.4.The content of sucrose,fructose,soluble total sugar,glucose and starch in the leaves of potato virus-free seedlings was the highest in 0.1mmol/L in ZnSO4 treatment and the highest in 0.5mmol/L in the treatment of MnSO4 except H3BO3.The highest is 0.2mmol/L.5.From the relationship between the growth index and several sugar contents of the virus-free seedlings and the concentration of NH4NO3,ZnSO4,MnSO4 and H3BO3,NH4NO3,ZnSO4,MnSO4 and H3BO3 were cultured in the culture process of the virus-free seedlings of Favorui.Suitable concentrations should be 20mmol/L,0.1 mmol/L,0.5mmol/L,and 0.4mmol/L,respectively.
