Effects Of Anti-Stress Agents On Liver And Duodenum Injury In Egg-Ducks

This exper iment was intended to explore add ing 4 kinds of anti-str ess agent-VC,VE,resver atrol(Res)and super ior putin on the cage jinyun hemp duck c linical s hape,blood biochemical indicators,antioxidant capacity,histomorphology,str ess related genes GRP78,COX2 and inf lammation changes of INOS m RNA and protein expr ession,by compar ing the f our kinds of the use effect of anti-str ess agent,pr eliminar y revealed the poss ible molecular mechanism of action,to explore the anti-str ess agent on the liver and duodenum oxidative stress injury of protection.Select ing 300 only 82 days of age's healthy jinyun sheldr ake,were randomly divided into control group(jinyun sheldr ake + bas e fodder),test A gr oup bas ed diet + vitamin C(500 mg/kg),test B group(basal diet + vitamin E 375 mg/kg),test C group based diet + resveratrol(500 mg/kg),test D group based diet + optimal putin(625 mg/kg),17 days of the tr ial per iod,could be divided into flat and caged two phases.Serum indicators related to liver function were detected: total protein,albumin,globulin,total bilirubin,AST,ALT,MDA,S OD,CAT,GS H-P x and total antioxidant capac ity.The antioxidant capac ity of t issue inc luded malondialdehyde,super oxide dis mutase and total antioxidant capac ity,total protein,glutathione peroxidas e and catalase.Pathological analys is of liver tissue and duodenum was detected by HE staining.MRNA tr anscr iption and protein express ion of COX2,GRP78 and INOS genes in liver and duodenum wer e detected by RT-PCR and Western Blot.The specific results are as follows:(1)By obs erving the clinical shape of laying ducks on the cage,it was found that laying ducks were in poor mental state after the cage,with incr eased body temperature,f ond of s leeping,less activity,incr eased body temperature,decreased appetite and egg-laying rate,depressed a nd unstable standing;Laying ducks gradually improved after the addition of four anti-stress agents.(2)Serum immune biochemical indexes and tissue antioxidant capac ity test indexes : T-AOC,MDA,GS H-Px and T-BI L contents f luctuated w ith the dur ation of caged str ess;I l-1,IL-6,AST and ALT showed a decreas ing trend.I L-4,TP,ALB,GLB,CAT and S OD showed a r is ing tr end.With the increas e of the time of caging str ess,the AST,ALT,TP,ALB,GLB,T-BI L,MDA,I l-1,IL-6 and other indicators increased after th e addition of four anti-stress agents vitamin C.In the antioxidant capac ity of tissues,TP,CAT,S OD,GS H-Px and T-AOC increased,while MDA content decreas ed.Group E showed th e most signif icant differ ence(P<0.05).VC,VE,resver atrol and optimal putin could s ignif icantly reduce ser um MDA,AST,ALT,TP,ALB,GLB,T-BIL,MDA,IL-1?,IL-6,TP content of pathogenies caged,increase SOD,GS H-Px and CAT,T-AOC content,which showed anti-stress agent had signif icant Kang S hang cage and played a role of immune stress ability,pathogenies signif icantly enhanc ed the immune function,and had certain effect to protect liver function.(3)Tissue str ucture: VC,VE,resver atrol and upr edine could s ignif icantly improve the liver and duodenum damage.According to the observation of histological s ection morphology,the anti-caged stress additive could change the liver and duodenum of the dis eas ed laying duck under the condition of relieving stress.Of the var ious additives,youpuding had the best anti-caging effect,followed by resveratrol,and finally vitamin.(4)The m RNA or protein expr essions of COX2,INOS and GRP78 genes in the liver of cage-stress ed laying ducks induced by VC were down-regulated and up-r egulated in the duodenum by RT-PCR and Western Blot.VE induced up-regulated m RNA or protein expr ession of COX2,INOS and GRP78 genes in liver and duodenum of caged stress laying ducks.Resver atrol and upredine could increase the m RNA or protein expr ession of COX2,I NOS and GRP78 genes in the duodenum and liver of the up per caged duck exposed to stress.These four anti-stress agents might inhibit the expression of genes such as oxidative stress and inflammatory factors.
