Symbiotic bacteria Wolbachia infected with a wide range of arthropods,and many mites are infected,Wolbachia play a important role in the host reproduction.Wolbachia can cause sustained and stable infection in the host,Wolbachia can be transmitted to generation through the cells of the host,including horizontal transmission and vertical transmission.The most common way that Wolbachia causes host reproductive abnormalities is to induce cytoplasmic incompatibility(CI).Although there are many studies on the CI mechanism,the mechanism of CI is still unclear.The distribution of Wolbachia in the population of spider mites is widespread.In this study,the Wolbachia strains infected with three spider mites were analyzed by PCR and multilocus sequence typing(MLST),the effect of Wolbachia on the reproductive regulation of the host was studied by biological hybridization.The results showed that the three spider mites infected with four different strains of Wolbachia,and all of them belong to the B supergroup,wTupe3 has a close relationship with wTtrl and distant relationship with wTpuel.Among the four different Wolbachia strains infected with spider mites,when the males infected with Wolbachia mating with uninfected females,causing different degrees of the Cl,wTpuel Cl is the highest,wTtr,wTur CI is weak,wTupe3 CI is the lowest.Therefore,the infection of Wolbachia in spider mite is more complicated,and the Wolbachia strains are very diverse and Wolbachia of different strains has different effects on the reproduction and suitability.The type ? secretion system(T4SS)is an effective pathway for the regulation of transport DNA/protein macromolecules into eukaryotic cells.The T4SS were found and studied in Agrobacterium tumefaciens,Bordetella pertussis,Helicobacter pylori and Legionella pneumophila.The type IV secretion system is the channel between bacteria and the host interaction.Bacteria can secrete DNA or proteins through the T4SS into host eukaryotic cells.In this study,the T4SS virB8-D4 operon of four Wolbachia strains was identified,in order to understand the interaction mechanism between Wolbachia and the host,we studied the conservation and function of Wolbachia T4SS by comparing the nucleic acid and amino acid.The results showed that the T4SS of four Wolbachia strains was present,and the structure of virB8-D4 operon was consistent with other Wolbachia strains,and it also was similar to the T4SS structure of near source of Wolbachia.And it is found that Wolbachia T4SS is very conserved,suggesting that it may be related to Wolbachia maintain a stable infection in the host and transfer it to the offspring,comparison of Wolbachia type ? secretion system flanking sequences ribA gene diversity,speculating that it may be concerned with different reproductions caused by different Wolbachia strains.For the first time,the T4SS study of spider mite Wolbachia was initiated which provided a theoretical basis for further study the interaction mechanism between Wolbachia and the host.
