In veterinary clinics,animals such as dogs and cats often show signs of depression,loss of appetite,and cognitive dysfunction.This phenomenon seriously affects the treatment and recov ery of diseased animals in veterinary clinics.Therefore,how to improve this mental state is a difficult problem that needs to be solved urgently in veterinary clinic.At present,in human medicine,studies have shown that low doses of ketamine have a goo d antidepressant effect.Ketamine is also an anesthetic commonly used by veterinarians.Therefore,this experiment mainly relies on ketamine anesthesia in experimental animals and low-dose ketamine after surgery to alleviate postoperative mental state problems.In this experiment,the mouse lipopolysaccharide was used to induce depression model.The optimal frequency of drug was explored by different frequency of drug use,and the optimal drug frequency was applied to the dog experiment to reveal the effect of low-dose ketamine on postoperative inflammatory factors and mental status in dogs.To clarify its effect on the improvement of postoperative mental state in dogs in veterinary clinics.In this experiment,a mouse experiment was first used to induce a d epression model in all mice by intraperitoneal injection of lipopolysaccharide.After successful modeling,the mice were divided into control group,medication once,medication three times and medication seven times.The control group was intraperitoneally injected with normal saline after the model was established;the low-dose ketamine was injected in the first group after 24 h of modeling;the low-dose ketamine was injected in the three groups after 24 h,3 d,and 7 d after the model was established;Low dose ketamine.Then,the model 24 h,3 d,7 d,and 14 d were selected to use the open field experiment,forced swimming experiment,and sugar water preference experiment.At the end of the behavioral experiment,the rats were sacrificed.The blood and brain tissues were used to detect the contents of BDNF,S100?,5-HT,GFAP and CORT,and the frequency of treatment with the best therapeutic effect was selected.After the dog experiment,the dog model was performed by laparotomy,and the drug was administered according to the optimal frequency of administration.Then the wound healing degree was observed 3 d before surgery,immediately after surgery,24 h,3 d,5 d,10 d and 14 d after surgery.And mental state,and blood,cerebrospinal fluid was used to detect BDNF,S100?,5-HT,GFAP,CORT content.The experimental results showed that in the mouse behavioral experiment,the consump tion of sugar water in the control group was significantly less than that in the medication group,and the consumption of sugar water was increased with time.In the frequency of medication,the seven-time treatment group was significantly better than the medication three times and the medication group;the forced swimming experiment showed that the control group showed weaker vigor and shorter struggle time.The seven-time group showed the strongest vitality and longer struggle time.The open field test showed that the control group showed weak excitability,fewer wearers and a single trajectory.The seven-time group showed strong excitability,more wearer and more complicated trajectory.Protein detection experiments showed that the GFAP,5-HT,S100?,and CORT values in the control group were higher,then gradually decreased,and the BDNF values were lower,then gradually increased,and the lifting effect was not obvious.The seven-time histone test showed a significant change in elevation.In the dog e xperiment,the dogs had no abnormalities before the operation and the diet was good.Postoperative mental state was low,loss of appetite,and then gradually returned to normal over time.Compared with the control group,the mental state and appetite recovery were faster,and the wound healing was better.Protein detection experiments showed that the GFAP,5-HT,S100?,and CORT values in the control group were significantly increased,and the BDNF values were significantly decreased,and then the preoperative level was slowly restored.The histidine test value was recovered faster than the control group.The experimental results show that low-dose ketamine can significantly improve the mental state by improving the levels of GFAP,5-HT,S100?,CORT,and BDNF in the blood brain.Therefore,veterinary clinic can use ketamine as a related therapeutic drug.
