The incidence rate of breast tumor is very high in the canine tumor disease,accounting for about 50%of the tumors in female dogs.Canine breast tumors are similar to human breast tumors in the etiology,histological origin and classification,so the natural canine is the best model for human breast cancer.Therefore,the study of canine breast tumor can not only provide a new direction for the prevention and treatment of small animal tumor disease,but also provide some reference for the study of human tumor.In this study,we collected the canine breast tumor samples from Nanjing area,and used the method of pathology to classify breast tumors,statistics the relationship between breast cancer and age and breed,and the proportion of benign and malignant tumors accounted for.We use gas chromatography for the determination of polyunsaturated fatty acid(PUFA)content,detection of canine breast tumor tissues using immunohistochemical method of cyclooxygenase 2(COX-2)and vascular endothelial growth factor(VEGF)expression.In order to study the changes of PUFA in canine normal breast tissues and tumor tissues,the relationship between the changes of the content of tumor and explore the PUFA related factors COX-2 and VEGF,and PUFA,COX-2,VEGF correlation analysis and canine mammary tumors,provide new methods and means for the prevention of canine breast tumor diagnosis and prognosis.1.This study shows that poodle is the highest incidence of canine breast cancer in 24 cases collected,and the highest incidence of middle-aged and older dogs over the age of 8,the average age at onset of the age of 10.2.2.24 cases of canine breast tumors were benign tumors,accounting for 25%,with a total of 18 cases of malignant tumors,accounting for about 75%of all cases.Types of benign tumors include fibroadenoma(4/6,66.67%)and fibroma(2/6,33.33%).And the type of malignant tumor including mixed breast cancer(5/18,27.78%)and invasive ductal carcinoma(1/18,5.56%),ductal papillary carcinoma(5/18,27.78%),undifferentiated breast cancer(1/18,5.56%),simple breast(4/18,22.21%)and breast cancer(2/18,11.11%).3.The contents of linoleic acid(LA)and total ω-6 PUFA in breast tumor tissues were significantly lower than those in normal tissues(P<0.05).The gamma linolenic acid(GLA),twenty-two six carbon acid(DHA)and total ω-3 PUFA was significantly higher than normal group(P<0.05),four arachidonic acid/linoleic acid(AA/LA)and AA/ω-6 PUFA is significantly higher than that of normal tissue(P<0.05).The ω-6 PUFA,AA/ω-6,AA/LA and ω-6/ω-3 PUFA in malignant tumor tissues were significantly higher than those in benign tumors(P<0.05),while ω-3 PUFA was significantly lower than that in benign tumors(P<0.01).4.The expression of COX-2 and VEGF were negative in normal breast tissue,while the positive expression rate of COX-2 in tumor tissues was 66.67%,and the positive rate of VEGF was about 66.67%.Analysis of the relationship between COX-2 and VEGF found a positive correlation between the expression of them.The relationship between COX-2,VEGF and PUFA was analyzed.The results showed that LA and ω-6 were negatively correlated with COX-2,AA/co-6,AA/LA and co-6/co-3 were positively correlated with COX-2.In summary,in 24 cases of canine breast cancer,breast cancer popular in the elderly aged over 8 dogs,and the proportion of malignant tumors was higher,which was mainly mixed breast cancer and ductal papillary carcinoma.In malignant breast tumors,the metabolism of ω-6 PUFA was more vigorous,and the expression of COX-2 and VEGF was high.COX-2 and VEGF can be used as one of the indicators to judge the canine breast tumor.COX-2 and ω-6/ω-3 can be used as a reference for the diagnosis of benign and malignant breast tumors.
