With the improvement of social economic level and living standard,the consumer demand of poultry meat has changed from quantity to quality.Therefore,the focus point of breeding researchers has transferred from improving production performance to meat quality.DNA methylation,molecular marker and transcriptome sequencing,etc were used to screen genes related to meat quality traits.Furthermore,RNA interference and gene overexpression were used to elucidate the function of them.Previous study showed that genomic DNA methylation of Wenshang barred chicken pectoral was significantly correlated with muscle fiber density and shear force.Meanwhile the specific methylation fragment of LRP1 was detected.The correlation between LRP1 and meat quality remained unknown,neither the regulation mechanism.Wenshang barred chicken were selected to analyze the correlation between the meat quality and LRP1 m RNA expression level.The results revealed that LRP1 was positively correlated with shear force and muscle fiber density(P<0.05),no significant correlation was found in other meat qualities.Muscle fiber characteristics are important factors of shear force,and tightly correlated with muscle fiber development.Meanwhile,the muscle fiber density is also one of the most important measurements in muscle fiber development.In following experiment,a chicken LRP1 RNAi vector transfecting in chicken embryonic myoblasts were constructed to detect the changes of genes related to muscle development,cell proliferation and apoptosis after LRP1 RNAi.The results showed that the expression levels of IGF-1,IGFBP-5,IGF-1R decreased significantly(P<0.05),IGF-2,Myo D decreased extremely significantly(P<0.01)and MSTN increased extremely significantly(P<0.01)respectively when LRP1 was subjected to RNA interference.Meanwhile the m RNA expression level of PMP22,CDKN2 C increased significantly(P<0.05)and p53 increased extremely significantly(P<0.01)whereas Bcl-x decreased significantly(P<0.05).The results indicated that LRP1 positively correlated with muscle fiber development.What's more,LRP1 can promotes the proliferation and suppression of apoptosis.Finally,the expression of LRP1 impact on meat quality.The results provide candidate genes in meat quality improvement and provide new ideas to explore the regulation mechanism under them.Meanwhile,the study can accelerate the improvement of meat quality which will lay a foundation of the breeding strategy of high yield and efficiency which based on high meat quality.
