The commercial cultivation of genetically modified crops has brought enormous economic benefits to human beings,and the cultivation of genetically modified crops has also attracted people's attention to the safety of ecological environment.An important part of the environmental safety assessment of genetically modified crops is the impact on natural enemies in the field.The Propylaea Japonica are the predatory natural enemies of cotton fields and are important indicator non-target insects for the safety evaluation of GM crops.In this paper,the safety of two new insecticidal proteins,Cry1 B and Cry1 Fa,against the Propylaea Japonica was studied.The main results are as follows:1.The effects of Cry1 B and Cry1 Fa proteins on the biological characteristics of the Propylaea Japonica were identified.There was no significant difference in developmental duration,emergence rate,and insect weight at different ages after feeding 0.5 mg/mL of Cry1 B and Cry1 Fa protein compared with the control.The results showed that the Cry1 B and Cry1 Fa proteins had no significant effect on the biological characteristics.2.The effect of the Cry1 B and Cry1 Fa proteins on the enzyme activity was confirmed.Compared with the control,after the Propylaea Japonica feeding 0.25 mg/mL,0.5 mg/mL of Cry1 B and Cry1 Fa protein,the activity of glutathione S-transferase(GST),catalase(CAT),peroxide and the enzyme(POD)had no significant effect;after feeding 1.0 mg/mL of Cry1 B protein,the activity of the glutathione-S-transferase(GST),catalase(CAT),peroxidase(POD)was significantly increased;the activities of catalase(CAT)and peroxidase(POD)were significantly increased after feeding 1.0 mg/mL of Cry1 Fa protein.3.The effects of Cry1 B and Cry1 Fa protein on detoxification and metabolism related genes were determined.Cry1 B protein and Cry1 Fa protein at different concentrations(0.25 mg/mL,0.5 mg/mL,1.0 mg/mL)were used to treat the 2nd and 3rd instar larvae of the Propylaea Japonica for 12 h,24 h,48 h.Afterwards,compared with the control,the expression levels of some detoxification and metabolism-related genes were not significantly affected,and the expression levels of some detoxification and metabolism-related genes were significantly affected.4.The effects of Cry1 B and Cry1 Fa protein on the symbiotic bacteria were determined.The dominant bacteria in the 2nd instar larvae were treated with Cry1 B and Cry1 Fa proteins of 0.25 mg/mL,0.5 mg/mL and 1.0 mg/mL respectively for 12 h,24 h and 48 h of the 2nd and 3rd instar larvae of the Propylaea Japonica.All were composed of Firmicutes,Proteobacteria,Bacteroidetes,Actinobacteria,Verrucomicrobia and Chloroflexi,which were consistent with the control group;the dominant phylum of the 3rd instar larvae was Firmicutes,Proteobacteria,Actinobacteria,Bacteroidetes,and Acidobacteria,which were consistent with the control group.
