Objective: To study the changes of serum metabolites in prenatal cold-stressed pregnant rats by liquid chromatography-mass spectrometry(LC-MS)metabolomics method,in order to reveal the prenatal cold stress to pregnant mothers and children from the perspective of metabolomics.The path of influence of generations provides a new scientific basis for finding effective measures to prevent prenatal cold stress by maternal intervention.Methods: Wistar pregnant female rats were used as the research model.The pregnant female rats were randomly divided into four groups: cold stress three-day group(CT),control three-day group(NT),cold stress seven-day group(CS),Control group of seven days(NS),10 per group.The CS group pregnant female rats were placed in the 4 ℃ artificial intelligence climate chamber for cold stimulation(cold stimulation for 7 days)on the 14 th day of pregnancy,and the CT group pregnant female rats were placed in the 4 ℃ artificial intelligence climate chamber for cold on the 18 th day of pregnancy.Stimulation(3 days of cold stimulation),the control group were kept in the environment of 22±2 ℃,and the four groups were fed with limited feeding and free drinking water.Blood collection was performed on day 21 of pregnancy and serum was separated.Then,the serum samples of each group were analyzed by LC-MS metabolomics method,and the differential metabolites in serum of pregnant mothers after cold stress were screened out.Results: Metabolomics showed that there were 19 metabolites in the CS-NS group(arachidonic acid,L-tryptophan,α-linolenic acid,palmitic acid,linoleic acid,etc.),CT-NT There are 23 metabolites in the group(arachidonic acid,L-glutamine,phospholipids,erucic acid,α-linolenic acid,etc.);the metabolic pathways involved include biosynthesis of unsaturated fatty acids,arachidene Acid metabolism,linoleic acid metabolism,glutamine and glutamate metabolism.The above results indicate that prenatal cold stress may affect maternal lipid metabolism,amino acid metabolism and synthesis and release of maternal oxytocin,aldosterone,luteinizing hormone,etc.,which may cause maternal inflammatory reaction,amino acid metabolism deficiency and endocrine.The risk of high incidence of related diseases affects the growth and development of offspring.Conclusion: Pregnancy mothers can affect the changes of serum metabolites after cold stress.There were 19 metabolites in the serum of pregnant female rats in the prenatal cold stress seven-day group.There were 23 metabolites in the serum of pregnant mothers during the prenatal cold stress three-day group;α-linolenic acid,cis-(6,9,12)Linolenic acid,linoleic acid,hippuric acid,4-mercaptobutyric acid,etc.are expected to serve as a marker metabolite for early warning of prenatal cold stress.
